甲型H1N1流感患者Th17细胞减少机制的探讨 被引量:3

摘要 目的探讨甲型H1N1流感患者Th17细胞下降的可能机制。方法 40名健康人、53名单纯甲型H1N1流感患者、16名并发肺炎甲型H1N1流感患者入选本研究,以ELISA方法测定三组人群血浆TGF-β、IL-6、IL-1-β、IL-22、IL-23、IFN-γ水平,实时定量RT-PCR方法测定三组人群PBMC中RORγt、STAT3、SOCS3表达水平。统计学方法采用单因素方差分析,统计软件采用Graphpad Prism 4.0。结果甲型H1N1流感患者血浆TGF-β水平较健康人显著下降,并发肺炎甲型H1N1流感患者血浆TGF-β水平进一步显著下降(F=32.42,P<0.0001)。单纯甲型H1N1流感及并发肺炎甲型H1N1流感患者PBMC中STAT3表达显著低于健康人(F=4.599,P=0.0176)。结论甲型H1N1流感病毒可能通过降低血中TGF-β水平抑制核转录因子STAT3的表达,从而抑制Th17细胞的分化。 Abstract:Object To investigate into the possible mechanism of reduction of Th17 cells in A (H1N1) influenza patients. Methods Forty health people,53 mild H1N1 inlfuenza patients and 16 severe H1N1 influenza patients were enrolled in this study. TGF-β ,IL--6,IL-l-13 ,IL-22,IL-23 and IFN-3,1evel in plasma were detected by EL/SA,and Iranscriptional factors ROR"/t,STAT3 and SOCS3 in PBMC were detected by real-time quantitative RT-PCR. data was compared by one-way ANOVA and performed by Graphpad Prism 4.0 software. Result The level of TGF- 13 in plasma of the mild H1N1 influenza patients was significantly decreased compared to the health people,Moreover,The level of TGF- 13 in plasma of the severe H1N1 influenza patients with pneumonia was even more significantly decreased compared to the mild H 1N 1 influenza(F=32.42, P〈0.0001 ).In addition, transcriptional factors STAT3 in PBMC of H 1N 1 influenza patients (both mild and severe) was significantly decreased compared to the health people (F--4.599,P=0.0176). Conclusions H1N1 influenza virus night involve in the reduction of GF- 13 level and it can inhibit the differentiation of Thl7 through reducing the level of TGF- 13 level and the expression of STAT3 .
