摘要
目的建立血清中甘氟的气相色谱-质谱检测方法。方法用乙酸乙酯萃取血清中甘氟,50℃氮吹浓缩后,气质联用仪直接测定,采用EI源选择离子模式(SIM)检测,基质标准外标法定量。结果甘氟Ⅰ在2μg/L^200μg/L,甘氟Ⅱ在1μg/L^100μg/L范围内线性良好,检出限分别为0.6μg/L与0.3μg/L,定量限分别为2.0μg/L与1.0μg/L。甘氟Ⅰ的回收率在91.3%~104.4%之间,RSD(n=6)为4.7%~9.7%;甘氟Ⅱ的回收率在90.7%~105.1%之间,RSD(n=6)为2.6%~7.2%。结论方法操作简便、准确,灵敏度高,适于甘氟中毒病人的快速诊断检测。
Objective To establish a gas chromatography - mass spectrometry method for determination of gliftor in serum. Methods After extracted with ethyl acetate and concentrated with nitrogen at 50 ℃, the gliftor in senum was determined by GC - MS with selected ion monitoring mode, and quantified with matrix - match standard solution. Results The calibration curves were linear in the range of 2μg/L - 200 μg/L for gliflor Ⅰand 1 μg/L- 100 μg/LL for glifto Ⅱ. The limits of detection were 0. 6μg/Land O. 3 μg/L, and that of quantitation were 2.0 μg/Land 1.0 μg/L. The recoveries were 91.3% - 104.4% and 90.7% -105.1%, and the RSDs( n =6) were4.7% -9.7% and2.6% -7.2%, respectively. Conclusion The method is simple, accurate, high sensitive and it is suitable for detection of gliftor in serum for toxicological purposes.
