摘要
目的应用中空纤维液相微萃取(HF-LPME)技术建立血液样品中氯丙嗪的气相色谱分析方法。方法对影响氯丙嗪HF-LPME的实验条件进行优化,优化的萃取条件为:采用聚偏氟乙烯中空纤维膜,以正辛醇为萃取溶剂,于搅拌速率为2500 rpm条件下在8 ml碱化后的样品溶液中萃取25 min,取1μl萃取液气相色谱进行分析。结果该方法的线性范围为0.01 mg/L^10 mg/L,最低检出限0.01 mg/L,富集倍数为12.6倍,相对回收率93%~102%,RSD9.2%~13.3%。结论建立的HF-LPME方法操作简单、高效、所需有机溶剂少,可避免残留和交叉污染,适用于血液中氯丙嗪的检测。
Objective A method for determination of chlorpromazine in blood samples using hollow fiber -liquid phase micro - extraction ( HF - LPME) coupled with gas chromatography (GC) was developed. Methods The extraction conditions of chlorpromazine in blood were optimized. The polyvinylidene fluoride hollow fiber membrane ( PVDF - HF) and n - caprylic alcohol were employed. The sample solutions were alkalified and extracted at a stirring rate of 2500 rpm for 25 min. Results The linear range for chlorpromazine in blood was 0.01 mg/L - 10 mg/L, the detection limit (LOD) was 0.01 mg/L, the enrichment factors was 12.6, the recovery was 93% - 102%, RSD were 9.2% - 13.3%. Conclusion The HF - LPME method is simple, efficient, requiring less solvent and can avoid the residue and cross contamination, it is suitable for extraction and detection of chlorpromazine in blood samples.
