In vitro antibacterial activity of doripenem

摘要 目的评价多利培南对我国近3年临床分离致病菌的体外抗菌活性及影响因素。方法最低抑菌浓度(MIC)测定用琼脂二倍稀释法。结果多利培南对革兰阳性菌、革兰阴性菌以及厌氧菌都具有很强抗菌作用。其对甲氧西林敏感葡萄球菌和链球菌的MIC值均≤1mg·L^-1,抗革兰阳性菌作用与亚胺培南相似,优于美罗培南。对革兰阴性菌,多利培南抗菌作用与美罗培南相似,优于亚胺培南。细菌接种量、培养基pH值及血清含量变化对多利培南抗肺炎链球菌和铜绿假单胞菌的抗菌活性基本无影响。对金葡菌的MIC值,随培养基pH值和马血清含量升高,可上升2—8倍。对大肠埃希菌的MIC值,也随马血清含量升高上升2—8倍。结论多利培南对我国近3年临床分离的致病菌具有很好的体外抗菌作用,是现有碳青霉烯类药物中抗菌作用最为均衡的一个品种。 Objective To evaluate the in vitro antibacterial activity of doripenem against clinical isolated pathogens from recent three years. Methods The MICs were determined by the twofold agar dilution meth- od according to CLSI standards. Results Doripenem exhibited excellent antibacterial activity against G^+, G^- and anaerobes. MIC values of doripenem against methicillin - susceptibility Staphylococcus spp. were ≤ 1 mg · L^-1. It's similar to imipenem and better than meropenem, Anti- bacterial activities of doripenem against Gram - negative bacilli were comparable to meropenem and greater than imipenem. There were no significant effects of inoculum size, pH and horse serum on MICs against Streptococcus pneumoniae and Pseudomonas aeruginosa. The MIC values of doripenem against Staphylococcus aureus were increased 2 -8 times with rising pH and horse serum. Agains'tEscherichia coli, the MIC values were increased 2 - 8 times with rising horse serum only. Conclusion Doripenem showed excellent antibacterial activity against clinical isolates in recent three years in China. It has a well - balanced wide -spectrum against gram -positive and -negative bacteria ancl cleserved further study.
