金针菇菌丝生物量与其DNA含量标准曲线的构建 被引量:3

摘要 构建了金针菇(Flammulinavelutipes)FV菌株菌丝生物量与其提取的DNA含量的标准曲线。通过液体传代培养金针菇FV菌丝体,发现所得菌丝体的DNA含量受到菌种传代次数的影响。在标准条件下独立培养i个批次金针菇FV菌丝体构建标准曲线具有很好的线性(R。〉0.999)和重现性,统计构建标准曲线的所有60个数据,测定的单点误差范围为0.09%~5.86%(〈6%)。采用相同的方法检测金针菇FV、G1、F29、F29-S-121、F29-S-143,几个菌株标准菌丝的DNA含量分别为36.49±0.83、34.20±0.90、22.15±0.54、19.30±0.87、19.49±0.76pgjmg,其中白色菌株(FV、G1)菌丝中DNA含量远高于其它黄色菌株。 Standard curves relating DNA content and mycelial biomass grown in submerged culture were constructed for Flammulina velutipes, strain FV. Although the frequency of subculture affected the mycelial DNA content, when standardized mycelium growth conditions and DNA extraction procedures were adopted, standard curves relating DNA concentration with biomass for three separate batches of mycelium exhibited a good linear relationship (correlation coefficient ~0. 999) and reproducibility. The error range for a total of 60 determinations (12 samples, five replicates of each sample) was 0.09% ~5.86%. Recorded mycelial DNA contents of five F. velutipes strains (FV, G1, F29, F29-S-121 and F29-S-143) determined under identical conditions were 36.5,34.2.22.2.19.3 and 19.5 μg DNA per mg (dry wt) of mycelium, respectively. The DNA content of strains producing white fruit bodies (FV and G1) was much higher compared with yellow fruit body-producing strains.
