摘要
目的:研究身痛逐瘀胶囊对小鼠中枢和外周镇痛作用及冰醋酸致痛小鼠血清中NO、SOD、MDA、PGE2的影响。方法:以冰醋酸致痛和福尔马林致痛动物为模型,研究身痛逐瘀胶囊的中枢和外周镇痛作用;以试剂盒和ELISA法测定冰醋酸致痛小鼠血清中NO、SOD、MDA、PGE2的含量。结果:身痛逐瘀胶囊1.64g/kg、0.82g/kg、0.41g/kg能显著减少小鼠的扭体次数;1.64g/kg、0.82g/kg能显著降低第一时相累积反应时间;1.64g/kg能显著降低第二时相累积反应时间。与模型对照组相比,身痛逐瘀胶囊1.64g/kg、0.82g/kg均可以显著降低血清中NO、MDA、PGE2含量、升高血清中SOD含量;0.41g/kg显著降低血清中NO、MDA含量、升高血清中SOD含量。结论:身痛逐瘀胶囊可预防冰醋酸和福尔马林诱导的疼痛而具有中枢和外周镇痛作用,其机制与下调血清中NO、MDA和PGE2的水平,提高SOD的水平有关。
To observe the analgesic effects of Shentongzhuyu capsules on the pain model of mice which induced by acetic acid and for malin and explore its mechanisms. Methods: Shentongzhuyu capsules was divided into three groups as follows: high dose ( 1.64g/kg), mid dle dose (0.82g/kg), and low dose (0.41g/kg)with lavage administration using ibuprofen sustained-release capsules and yuanhuzhitong tablets as model drugs. Acetic acid writhing method and formalin-induced pain were used to study analgesic effect of Shentongzhuyu capsules. Determine the level of NO, SOD, MDA and PGE2 in serum of each group to observe the effect of Shentongzhuyu capsules on acetic acid-in duced pain. Results: In comparison with the model controls, at the doses of 0.41 ~ 1.64g/Kg of Shentongzhuyu capsules could reduce the number of writhes caused by acetic acid-induced writhing and at the doses 1.64g/kg, 0.82g/kg could decrease the reaction time of the first and the second phase of formalin-induced pain. High dose (1. 64g/kg)and middle dose(0.82g/kg) of Shentongzhuyu capsules can markedly decrease the level of NO, MDA and PGE2, improve the level of SOD in serum of pain model mice compared with the model group( P 〈 0.05, P 〈 0.01 ). Low dose (0.41g/kg) can markedly decrease the level of NO and MDA, improve the level of SOD in serum of pain model mice compared with the model group (P 〈 0.05, P 〈 0.01 ). Conclusion: Shentongzhuyu capsules has showed significant analgesic effect, connect ed with improving central and peripheral pain. Shantongzhuyu capsules may prevent and relieve pain induced by acetic acid effectively through reducing the level of NO, MDA and PGE2, improve the level of SOD in serum.
