摘要
目的初步探讨愈骨胶囊(血竭、骨碎补、续断、自然铜和延胡索)对新西兰兔桡骨骨折愈合的分子机制。方法将实验动物随机分成正常组、模型组、愈骨胶囊组和接骨七厘片组,柴本甫法造成骨折模型,于给药第2、4、6周进行生物力学测定,HE、Masson、甲苯胺兰染色观察骨痂组织病理变化,免疫组化观察愈骨胶囊对骨痂组织中转化生长因子-β(TGF-β),血管内皮生长因子(VEGF)的影响。结果与模型组相比,在第4、6周,愈骨胶囊组最大载荷,最大位移和弹性模数显著增加,有统计学差异(P<0.05);愈骨胶囊组软骨细胞增生明显、数目较多,软骨骨痂形成较早,钙化提前,纤维性骨痂、软骨岛、编织骨及皮质骨出现均较模型组早,骨折端塑形改造良好,髓腔再通时间缩短;在骨折后第4周时,愈骨胶囊组骨痂中VEGF、TGF-β的表达明显较模型组强。结论愈骨胶囊能使骨痂生长、塑形提前,促进骨折愈合,可能与提高骨痂组织内VEGF、TGF-β表达,完善骨痂内微血管重建有关。
AIM To explore the mechanism in promoting radial fracture healing in rabbit induced by Yugu Capsules. METHODS The experimental rabbits were randomly divided into 3 groups: blank control group, Yugu Capsules group and Jiegu Qili Tablets group. The models with 3 mm bone defect were produced in the middle of the right radius in rabbits. In the 2nd , 4th , 6th weeks after operation, the biomechanics were determined and the patho- logical change of the callus was observed after dying by HE, Masson, and methylaniline respectively to evaluate the healing of fracture. TGF-β and VEGF in the callus were studied to explore the mechanism in promoting radial fiacture healing. RESULTS Comparing with the blank control group, the values of maximum load, maximum displacement and elastic modulus were significantly higher in the 4th, 6th week after the administration of Yugu Cap- sules (P 〈 0.05) ; the proliferation of the chondrocyte was obvious, the formation of cartilage callus, fibrous cal- lus, cartilage islands, woven bone and cortex bone were earlier, the remolding of bone fracture and the recanalization of cavum medullare were better than those in the blank control group. In the 4th week, the expressions of VEGF, TGF-β in the callus of the Yugu Capsules group were obviously higher than those in the blank control group. CONCLUSION The mechanism of accelerating bone fracture healing by Yugu Capsules in rabbits may be associated with the increase of expression of VEGF, TGF-β, then promoting reestablishment of the capillary vessel in callus.
