摘要
目的 观察侯氏黑散方中风药、补虚药对脑缺血大鼠侧脑室水通道蛋白-4(AQP-4)及胶质纤维酸性蛋白(GFAP)表达的影响.方法 线栓法阻断大鼠大脑中动脉,制备局灶性脑缺血大鼠模型,分别在脑缺血6、24、48 h观察侯氏黑散方中风药、补虚药、风药补虚药联用对GFAP及AQP-4表达的影响.结果 脑缺血6 ~48 h,模型组大鼠侧脑室星形胶质细胞明显肿胀,GFAP、AQP-4表达较假手术组明显增强(P<0.01).脑缺血6h,风药、联用药可明显下调AQP-4表达,与模型组相比差异显著(P<0.05);脑缺血24 h,补虚药、联用药大鼠侧脑室AQP-4表达较模型组明显降低(P<0.05);脑缺血48 h,风药可明显降低GFAP表达,补虚药可明显下调GFAP、AQP-4的表达,与模型组相比有显著性差异(P<0.05).结论 风药、补虚药可通过调控GFAP、AQP-4表达,减轻星形胶质细胞水肿,对缺血性脑损伤具有积极的保护作用.
Objectives To observe the influences of wind-dispelling and tonifying Chinese medicinals in Houshihei San on the expressions of glial fibrillary acidic protein (GFAP) and aquaporin-4 (AQP-4) in lateral ventricle in rats with cerebral ischemia.Methods The model of focal cerebral ischemia was established by using thread approach to occlude middle cerebral artery in rats.The effects of winddispelling,tonifying and combining Chinese medicinals in Houshihei San on expressions of GFAP and AQP-4 were observed at three time points (6 hours,24 hours and 48 hours after cerebral ischemia).Results After ischemia for 6 hours to 48 hours,astrocytes in lateral ventricle were markedly swollen in model group,and expressions of GFAP and AQP-4 increased significantly in model group compared with those in sham-operation group (P < 0.01).After ischemia for 6 hours,wind-dispelling and combining Chinese medicinal significantly regulated downwards AQP-4 expression,which showed a significant difference compared with that in model group (P < 0.05).After ischemia for 24 hours,AQP-4 expression in lateral ventricle decreased significantly in tonifying group and combining group compared with that in model group (P < 0.05).After ischemia for 48 hours,wind-dispelling Chinese medicinal reduced significantly the expression of GFAP,and tonifying Chinese medicinals significantly regulated downwards the expressions of GFAP and AQP-4,which showed a significant difference compared with those in model group (P < 0.05).Conclusion Wind-dispelling Chinese medicinal and tonifying Chinese medicinal have positive protective effects on ischemic brain by controlling the expressions of GFAP and AQP-4 and reducing edema of astrocytes.
