补阳还五汤对实验性脑缺血大鼠水通道蛋白-4表达的影响 被引量:7

摘要 目的:观察补阳还五汤对大鼠脑缺血后AQP-4表达和患肢恢复功能的影响。方法:100只SD大鼠分为损伤组和中药组,均手术致脑缺血。中药组术后即给予补阳还五汤口服液灌胃;损伤组给予等量蒸馏水灌胃。术后1、3、7、14及21天分别对大鼠进行BBB评分和斜板实验检查患肢功能,然后处死,应用免疫组织化学技术检测脑组织AQP-4的表达。结果:脑缺血后第1天,两组受损伤脑灰质、白质中可见到AQP-4的表达明显增加;第3天时达到高峰,损伤组更明显(P<0.05);第7、14及21天,中药组AQP-4表达的减少几乎与神经功能的改善平行,与损伤组比较差异有非常显著性(P<0.01)。结论:补阳还五汤可能通过抑制脑缺血后AQP-4表达,消除脑水肿,减轻脑损伤,从而使残存脑组织保存并促进肢体功能恢复。 We investigated the effects of Buyanghuanwu Tang formula on AQP-4 expression in ischemic rats' brain, and associated functional recovery of limbs. In the study, 100 SD rats with incomplete ischemic brain were divided into injury group and traditional Chinese medicine group. The TCM group received Buyanghuanwu Tang formula after operation, and the injury group are fed with distilled water. BBB scale and inclined plane tests were performed to see the functionality of the limbs. The animals were sacrificed 1, 3, 7, 14 and 21 days after surgery. The expression of AQP-4 in brain was measured using immunohistochemical method. We found that the expression of AQP-4 was significantly increased in the gray and white matter area of brain one day after ischemia, and reached the peak on day 3 in both groups. Meanwhile, the injury group has seen a more significant increase (P〈0.05). The increase of AQP-4 expression in the TCM group ran parallel with the improvement of nerve function on the 7th,14th and 21st day. Comparing with the injury group, the expression of AQP-4 in TCM group was significantly lower(P〈0.01). We believe that Buyanghuanwu Tang formula eliminates the edema of brain, and alleviates the successive ischemia by inhibiting the expression of AQP-4 after ischemia, playing a role of protecting unhurt brain tissues, and facilitating the recovery of limb functions.
