伤科接骨片促进实验性骨折愈合的超微结构观察 被引量:24

摘要 以实验性骨痂组织的超微结构变化为依据 ,进一步探讨伤科接骨片促进骨折愈合的机理。取 2 0只健康雄性昆明种小鼠 ,造成左桡骨中段 1.5 mm缺损骨折模型。将模鼠随机分为 2组 :实验组小鼠喂服伤科接骨片煎剂每只每天 2 .5 ml(含生药 5 g) ;对照组小鼠喂服等体积的生理盐水。两组小鼠在术后 2周及 4周分两批处死 ,取出骨断端骨痂组织 ,制成超薄切片 ,在日立 H- 6 0 0透射电镜下观察。结果示术后 2周 ,对照组小鼠骨痂内毛细血管少 ,内皮细胞功能不活跃 ,并有较多的成软骨细胞 ;实验组毛细血管多 ,管腔内有红细胞通过 ,内皮细胞器丰富 ,并出现大量的成骨细胞。术后 4周 ,对照组小鼠骨痂内毛细血管增多 ,增粗 ,并出现许多成骨细胞 ;实验组有大量的骨细胞形成。表明伤科接骨片能够促进骨折愈合。推测其作用机理为 :1促进毛细血管再生 ,改善局部微循环。 2促进成骨细胞的分化及增殖。 3补充骨折愈合所需微量元素。 4诱导成骨。 In order to study the mechanism of promoting fracture healing by “Shangke Jiegu ”Tablet based on the ultrastructure, 1.5mm defect fractures in left radial middle shafts were made in 20 healthy mice. The mice were randomly divided into 2 groups:experimental group(A) were given“Shangke Jiegu ”Tablet with a daily dosage of 2.5ml every mouse(containing crude drugs 5g),while control group(B) were given the same volume of normal saline. The animals of two animals were sacrificed afte 2 and 4 weeks respectively,the callus were taken out to be observed by transmission electron microscope. Result:2 weeks after operation,the capillaries in B were fewer than that of A,and the endothelical cells were not more active,there were more chondroblasts in B, while in A there were more capillaries, and there were erythrocytes passing through the lumen rich endothelial cell and a lot of osteolasts. 4 weeks after operation, the capillaries increased and became coarser in B, and the inactive osteoblasts were seen, while in A, the osteocytes and bone matrix miniralization were seen. The result showed that“Shangke Jiegu”Tablet can promote fracture healing, and it's mechanism may be: ① Promoting capillary regeneration and improving local microcirculation. ② Promoting differentiation and hyperplasia of osteoblast. ③ Supplying the needed microelement of fracture healing. 4 Inducing osteogenesis.
