摘要
【目的】比较安寐汤不同极性部位的镇静催眠作用。【方法】依次使用石油醚、乙酸乙酯、正丁醇提取安寐汤原方水提物,获得不同的极性部位,通过经典中枢神经系统镇静催眠药理实验,以入睡率、入睡潜伏期、睡眠维持时间为指标,比较各极性部位的镇静催眠作用。【结果】安寐汤乙酸乙酯部位和正丁醇部位能显著抑制小鼠的自主活动;对戊巴比妥诱导的睡眠表现出协同作用,不仅显著增加了催眠剂量戊巴比妥小鼠的入睡只数,而且能显著延长睡眠维持时间,增加阈下剂量戊巴比妥钠睡眠小鼠个数,具有一定的镇静、催眠作用。【结论】安寐汤乙酸乙酯部位和正丁醇部位为其镇静催眠作用的活性部位。
Objective To study the sedative and hypnotic effects of active constituents of different polarity extracted from Anmei Decoction on mice. Methods The constituents of different polarity were extracted by petroleum ether, ethyl acetate and n-butanol, respectively. Sleep rate, sleep latency and sleep duration were observed by the classic sedative and hypnotic pharmacological experiments for central nervous system. Results The results showed that the constituents extracted from Anmei Decoction by n-butanol and ethyl acetate significantly inhibited the mice autonomic activities and had the synergic effect on sleep with pentobarbital, which not only increased the number of mice falling asleep in hypnotic dose of pentobarbital, but also prolonged the sleep time and increased the number of mice falling asleep in subthreshold dose of pentobarbital. Conclusion The constituents extracted from Anmei Decoction by n-butanol and ethyl acetate are the active fractions with sedative and hypnotic activities.
