摘要
采用RT-PCR和RACE法从超旱生、耐盐植物梭梭(Haloxylon ammodendron)中扩增出VHA-B基因cDNA序列,其开放阅读框为1 413bp,推测编码全长为471个氨基酸残基的氨基酸序列,该蛋白分子量约为52.304ku,等电点为5.08。运用SMART、DNAMAN、GOR4、TMPred等生物信息学软件对梭梭VHA-B功能域、跨膜结构进行分析显示,梭梭VHA-B含有3个ATP-synt_ab_N、ATP-synt_ab、ATP-synt_ab_C结构域,但是不含跨膜结构域和信号肽。氨基酸序列同源性结果显示,HaVHA-B与盐爪爪、盐穗木、碱蓬等同源性高达89.68%。说明VHA-B基因是一个高度保守基因。
Through RT-PCR(reverse transcription polymerase chain reaction)and RACE(rapid ampli- fication of cDNA ends), a 1 413 bp fragment containing entire VHA-B coding region has been ob- tained from the extreme xerophyte and salt-tolerant plant Haloarylon arnmodendron. The cDNA codes a protein with 471 amino acids, and the molecular weight is about 52. 304 ku, isoelectric point is 5.08. Based on the analysis of bioinformatics software, such as SMART, DNAMAN ,GOR4 and TMPred, Haloa:ylon ammodendron VHA-B contains three domains, including ATP-synt ab_N, ATP-synt_ab, and ATP-synt_ab_C, but does not contain trans-membrane domain and signal peptide. The similarity comparison result of deduced amino acid sequence indicated that the HaVHA-B shared 89. 68% sequence identity with published homologous of the species such as Kalidium foliatum, Halostachys caspica and Suaeda corniculata, which indicated that VHA-B gene was a highly conserved gene.
