摘要
目的 :提取高纯度的甲状腺球蛋白 (Tg) ,为制备Tg 抗血清及Tg单克隆抗体 ,建立Tg的检验方法及TgAb的测定方法做准备。方法 :将手术切除的新鲜人甲状腺组织 ,加入生理盐水匀浆、过滤 ,把滤液浓缩 ,经二次SephadexG -200柱层析 ,再经DEAE纤维素离子交换层析。结果 :获得较高纯度的人Tg ,经SDSPAGE鉴定 ,显示为1条区带 ,表明纯度较高。经免疫电泳鉴定 ,提纯的Tg有较好的免疫活性。结论 :这种提取Tg 的方法 ,简便易行 ,分离效果好 ,适用于大量制备抗原。
Objective: To establish the immunoassay of thyroglobulin (Tg) and anti-Tg antibody (TgAb), with use of Tg antiserum and Tg monoclonal antibody by immunization of highly purified Tg. Methods: A Surgical specimen of human thyroid tissue was homogenized in normal saline, filtered and then concentrated. The homogenate was purified on a colamn of Sephadex G-200 twice and further passed through a DEAE cellulose column. Results:Highly purified Tg was obtained. Single band from purified Tg was found in SDS-PAGE, indicating that the purity of Tg was high. The immune activity of purified Tg kept well by immune electrophoresis identification. Conclusion: The method for preparing Tg can be used for large production of Tg antigen with respect of performing easily and isolating completly.
