摘要
目的:建立大鼠血浆中棉酚含量的高效液相色谱测定方法。方法:采用ODS-C18色谱柱(4.6 mm×200 mm,5μm),柱温25℃,流动相为甲醇-1%磷酸溶液(85∶15),流速1.0 ml/min,检测波长:235 nm。结果:在0.33~3.30μg/ml范围内峰面积与浓度呈现良好的线性关系(r=0.999 4),检测限为0.04μg/ml;平均回收率为94.7%~97.3%;给药组大鼠血浆中棉酚的含量为0.68~2.14μg/ml。结论:该方法可用于棉酚生物样品分析。
Objective :To establish an HPLC method for determination of gossypol in rat plasma. Methods :An ODS -C18 column (4.6 mm ×200 mm, 5 μm) was used as stationary phase, and methanol and 1% phosphoric acid solution (85: 15, v/v) as mobile phase with the flow rate of 1.0 ml/min. The detective wavelength was at 235 nm. Results :The peak area of gossypol and its concentration showed a good linear relationship when the concentrations varied from 0.33 - 3.30 μg/ml, related coefficient being 0. 999 4. The limit of detection was 0.04μg/ml ( S/N = 3 ). The relative recovery was between 94.7% and 97.3%. The content of gossypol in rat plasma was between 0.68 μg/ml and 2.14μg/ml. Conclusion:This method can be used for analysis of gossypol in biological samples.
