摘要
目的观察人第10号染色体缺失的磷酸酶及张力蛋白同源基因(phosphatase and tension homolog deteted from chromosome 10,PTEN)、iNOS、COX-2在大肠癌中的表达及相互关系,探讨三者在大肠癌发生、发展及转移过程中的作用。方法采用免疫组化SP法检测58例大肠癌中PTEN、iNOS、COX-2的表达,分析PTEN与iNOS、COX-2表达之间的相关性和临床意义。结果 (1)大肠癌组织中PTEN的阳性率为46.6%(27/58),显著低于正常大肠组织(P<0.01);iNOS、COX-2阳性率分别为79.3%(46/58)、79.3%(46/58),显著高于正常大肠组织(P<0.01)。(2)PTEN表达和淋巴结转移相关(P<0.05),与患者的性别、年龄、分化程度、肿瘤大小及浸润深度差异无统计学意义。(3)大肠癌组织中PTEN的表达与iNOS表达之间呈负相关(P<0.05),PTEN的表达与COX-2表达之间亦呈负相关(P<0.05)。结论 PTEN与iNOS、COX-2在大肠癌发生、发展过程中起重要作用,PTEN基因表达的缺失可能会上调iNOS、COX-2的阳性率,对肿瘤血管的形成产生协同作用,从而增加大肠癌发生转移的机率。
Purpose To examine association of PTEN with iNOS, COX-2 in human colorectal carcinoma and their relationship with tumor angiogenesis, clinieopathological features and prognosis. Methods Immunohistochemical technique was used to detect expres- sions of PTEN and iNOS, COX-2 in 58 cases of human coloreetal carcinoma. The association between PTEN and iNOS, COX-2 in tumor angiogenesis was analyzed. Results ( 1 ) Positive expression rates of PTEN in 58 cases of colorectal carcinoma tissue was 46. 6% (27/58), which was significantly lower than that in normal colorectal tissues (P 〈 0. 01 ). Positive expression rates of iNOS, COX-2 were 79. 3% (46/58), 79.3% (46/58), which were significantly higher than that in normal colorectal tissues (P 〈 0. 01 ). ( 2 ) The expression rates of PTEN was significantly associated with lymph node metastasis ( P 〈 0. 05 ), and not with age, sex, invasive depth, tumor size and tumor grading. (3) There were negative correlations among PTEN expression with iNOS, COX-2 ( P 〈 0. 05 ). Conclusion PTEN, iNOS, and COX-2 may play very important roles in the development of colorectal carcinoma. The loss of expres- sion of PTEN may induce tumor angiogenesis and invasion by activating the transcription of iNOS and COX-2.
