摘要
目的 研究血管内皮生长因子 (vascular endothelialgrowth factor,VEGF)反义寡核苷酸 (ODN )抑制人肝癌细胞系 VEGF的表达 ,探讨肝癌基因治疗的新方法 .方法 人肝癌细胞系 772 1能表达 VEGF,采用人工合成反义、正义 ODN分别作用于 772 1细胞 ,免疫组化图像分析方法比较各处理组VEGF的表达情况 ,即可了解 VEGF反义 ODN抑制肝癌的血管形成情况 .结果 VEGF反义 ODN确有抑制人肝癌细胞 VEGF表达的作用 .结论 VEGF反义 ODN能够抑制肝癌血管的形成 ,其有望成为抗肝癌的新一代基因治疗药物 .
AIM To investigate the possibility whether the antisense oligodeoxynucleotide (ODN) of vascular endothelial growth factor (VEGF) can become a new gene therapeutic drug for antiangiogenesis of human hepatocellular carcinoma (HCC). METHODS 7721 cell can express VEGF. Synthetic antisense ODN complementary to one region of VEGF mRNA was added into the medium of 7721. Then by means of the image analysis, it was determined whether the expression of VEGF of 7721 could be inhibited by the ODN. RESULTS Expression of VEGF of 7721 was inhibited by the VEGF antisense ODN. CONCLUSION It is promising that VEGF antisense ODN will become a new gene therapeutic drug for antiangiogenesis of HCC.
