摘要 用膜片钳单通道技术 ,观察在细胞膜内侧加入甲基莲心碱 (Nef)后 K+通道特性的改变 ,以研究Nef对豚鼠心室肌细胞及猪冠状动脉平滑肌细胞K+通道的作用 .结果 :Nef可使心肌细胞及冠脉平滑肌细胞上 K+通道开放概率 (PO)降低 . 1 0 ,2 0μmol· L-1Nef分别使心肌细胞 K+通道降低 (50± 32 ) %和 (96± 5) % ,30 ,50 μmol· L-1的 Nef分别使猪冠脉平滑肌细胞上 K+通道 PO 降低 (51±2 1 ) %和 (71± 1 4) % .结果表明 ,Nef可从通道内口阻断心肌及平滑肌细胞上 K+通道 。 To study the effects of neferine (Nef) on K + channels in guinea pig ventricular cells and porcine coronary artery smooth muscle cells (SMC), the changes of the properties of single K + channel after application of Nef were investigated using inside out patch clamp technique. Results indicated that Nef involved a blockade of K + channel both in guinea pig ventricular cells and porcine coronary artery SMC. Application of Nef 10, 20 μmol·L -1 from inner side of membrane of ventricular cells decreased the open probability(P O) of the channel by (50±32)% and (96±5)%, respectively, whereas in coronary artery SMC 30 and 50 μmol·L -1 Nef decreased P O of K + channel by (51±21)% and (71±14)%, respectively. This study suggests that Nef block K + channel from inner mouth of the channel both in ventricular cells and coronary artery SMC, and the blocking potency in the former be stronger than in the later.
