猪IGF-1及其受体基因部分组织表达规律的研究 被引量:6

Research on the tissue expression patterns of IGF-1 and IGF-1R in pigs

摘要 小型猪矮小机理的研究已经成为研究热点。胰岛素样生长因子-1是机体生长、发育和代谢的重要调控因子,主要通过与其受体的结合发挥作用。本研究以我国特有的版纳微型猪和西藏小型猪为主要研究对象,以大型猪种军牧猪和大白猪为对照,采用荧光定量PCR研究IGF-1及受体在心脏、肝脏、肾脏和肌肉组织中的表达规律。结果显示:小型猪中,这4个基因均在肝脏组织中表达量最高,肾脏组织其次,肌肉最低;大型猪中,IGF-1基因在肝脏中表达量最高、肌肉其次;IGF-1在大白猪肌肉组织中表达量最高,版纳微型猪中最低;IGF-1R基因在肾脏表达量最高、肌肉最低。综上,推断IGF-1及其受体基因可能是导致小型猪矮小的功能基因,本研究为小型猪的生长发育机制研究提供了分子依据。 Until now,the dwarfism mechanism for the miniature pigs as become a hot spot in the development area. Insulin like growth factor 1 (IGF-1), which binds with IGF-1R, plays crucial roles in the control of embryonic and postnatal development in the vertebrate species. In our re search, we analyzed the tissue expression patterns(in the heart, liver, kidney and muscle tissues) of IGF-1 and IGF-1R in the Banna miniature pig(BNMP) and Tibetan miniature pig compared with "Junmu" white pig and large white pig by real-time quantitative PCR. The results showed that the IGF-1 was expressed in the liverthe kidneythe muscle in the miniature pigs. In the large pigs, IGF-1 gene expression was the highest in the liver, next the muscle; the highest expression level of IGF-1 was found in the muscles of large white pig,and the lowest in BNMP. IGF-1R expression was the highest in the kidney,and the lowest in the muscle. From the above ail,IGF-1 and its re ceptor genes may be a gene family related with dwarfism. Our research laied certain molecular the oretical basis for the growth and development mechanism of pigs.
