摘要 目的探讨阿托伐他汀对老年亚临床动脉粥样硬化患者单核细胞环氧化酶2活性的影响。方法采用高分辨超声检测技术,筛选在我院体检的老年亚临床动脉粥样硬化患者48例(亚临床组),并选择体检正常者48例(对照组)。采用逆转录聚合酶链反应的方法,检测所有研究对象外周血单核细胞环氧化酶2的表达,同时观察亚临床组单核细胞在不同浓度阿托伐他汀干预24h后环氧化酶2表达的变化。结果亚临床组空腹血糖、收缩压、舒张压、LDL-C、吸烟指数、环氧化酶2mRNA表达较对照组明显增高,HDL-C较对照组明显降低,差异有统计学意义(P<0.05,P<0.01)。不同浓度阿托伐他汀明显抑制老年亚临床动脉粥样硬化患者外周血单核细胞环氧化酶2mRNA表达,且呈浓度依赖性(P<0.05)。结论阿托伐他汀通过降低外周血单核细胞环氧化酶2的表达,在老年亚动脉粥样硬化患者发挥调脂外的抗炎作用。 Objective To study the effect of atorvastatin on activity of cyclooxygenase 2 (COX-2) in mononuclear cells of elderly subclinical atherosclerosis patients. Methods Forty-eight elderly subclinical atherosclerosis patients diagnosed with high frequency ultrasonic technology served as a subclinical atherosclerosis group and 48 healthy subjects served as a control group in this study. Expression of COX-2 in peripheral mononuclear cells was detected by RT-PCR before and 24 h af- ter atorvastatin at different concentrations. Results The fasting blood glucose level,diastolic and systolic blood pressure, smoking index and COX-2 mRNA expression level were significantly higher whereas the LDL-C level was significantly lower in subclinical atherosclerosis group than in control group (P〈0.05 ,P〈0.01). Atorvastatin at different concentrations significantly inhibi- ted the expression of COX-2 mRNA in peripheral mononuclear cells of elderly subclinical athero- sclerosis patients in a concentration-dependent manner (P〈0.05). Conclusion Atorvastatin plays a role in regulation of lipid and antiinflammation in elderly subclinical atherosclerosis patients by reducing the expression of COX-2 mRNA in peripheral mononuclear cells.
