摘要
为了研究桉树种植地土壤真菌群落多样性,该试验利用ARDRA和ITS系列分析相结合的方法,提取桉树土壤微生物DNA,构建真菌的ITS克隆文库进行阳性克隆检测,分析显示含有ITS序列的阳性克隆子95个,并归类为13个不同的类群OTUs,随机挑取各类群进行测序,构建系统发育进化树.结果表明:用限制性内切酶HinfⅢ进行酶切鉴定,检测到13种不同的分类操作单位(OTUs),其中有3种为单一基因型(OTUs),占总数的23.1%.根据库容(coverage)公式计算得出文库的库容值为97.5%.采用ARDRA酶切图谱和测序结果分析,桉树人工林地土壤真菌群落由Ant fungus garden metagenome,Freshwater metagenome,Coral metagenome,Marine metagenome 4大类群组成,这为进一步研究桉树人工林土壤微生物及环境质量的变化提供了基础资料,并为西南地区桉树与土壤微生物数量、群落之间的变化关系提供了可靠的依据.
In order to research the diversity of the Eucalyptus plantation soil microbial fungi, we, in this experiment, have extracted microbial DNA in the Eucalyptus plantation soil by means of the ARDRA and ITS series Analysis, and tested positive clones by ITS clone libraries. The research shows that there are 13 different operating units (OTUs) by the restriction enzymes Hinf HI , and there are 3 kinds of for a single gene type (OTUs), occupied 23.1% of the total. According to the capacity of (calculated coverage) for- mula, the capacity of value is 97.5%. Through ARDRA enzymes map and sequencing result analysis show that fungus composed from Ant fungus garden metagenorne , Freshwater rnetagenome , Coral metagenome , and Marine rnetagenome in soil of Eucalyptus artificially woodland. As species are relatively scarce, this study is to provide theoretical basis for analysis for further study of the change of Eucalyptus plantation soil microbes, environmental quality, and the microbial quantity and community of microbial of Eucalyptus and soil.
