摘要
目的:以内毒素休克拟感染性休克,探讨补气活血解毒法在防治内毒素休克脑损伤中的保护作用与相关机制。方法::96只SD大鼠随机分为假手术组(生理盐水20 ml·kg^(-1))、模型组(脂多糖8 mg·kg^(-1))、加味生脉饮注射液组(脂多糖8mg·kg^(-1)加味生脉饮注射液10ml·kg^(-1))、地塞米松组(脂多糖8 mg·kg^(-1)+地塞米松5 mg·kg^(-1))等4组,每组再分为1,2,3,6 h 4个观测时点。各动物手术分离颈总动脉并与生物机能系统连接,监测平均动脉压(MAP),测定脑组织含水量、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性等指标,观察脑组织结构的变化。结果:模型组动物在注射脂多糖后MAP进行性下降,脑含水量百分数明显增加至(82.69±0.28)%(P<0.05),MDA含量明显增加至(9.02±1.19)nmol·mg^(-1)(P<0.05),SOD活性明显下降至(37.15±2.92)U·mg^(-1)(P<0.05),同时观察到脑微血管水肿,各治疗组上述病理改变明显改善。结论:生脉饮加味方有较好的抗休克、保护脑组织的作用,补气活血解毒治法可作为感染性休克中后期的中医治法。
Objective: To study the protection and related mechanism of promoting blood flow and deintoxication method in the treatment of brain injure in rats with septic shock imitated by endotoxin shock. Method : Totally 96 SD rats were randomly divided into 4 groups, the sham operated group (saline 20ml · kg-1) , model group (lipopolysaccharide LPS 8rag · kg-1 ) , reforming Shenmaiyin group ( LPS 8mg · kg-' plus Shenmaiyin 10ml · kg-1 ) , and dexamethasone group ( LPS 8rag · kg-1 plus dexamethasone 5mg · kg-1 ) , and each group was subdivided into 1,2,3h and 6h point. All animals were operated to separate carotid artery and connected bi- ological functional systems to monitor mean arterial pressure(MAP) , and the moisture, the content of malonaldehyde(MDA) , the ac- tivity of erythrocuprein(SOD) and the structural changes of brain tissue were determined as well. Result: MAP in the model group was decreased after the injection of LPS with water content increased to ( 82.69 ± 0.28 ) % ( P 〈 0.05 ) and the content of MDA increased to ( 9.02 ± 1.19 ) nmol · mg-1 ( P 〈 0.05 ). SOD activity was decreased to ( 37.15 ± 2.92 ) U mg- 1 ( p 〈 0.05 ). Capillary vessel edema in brain tissue was observed as well, and the above-mentioned pathological alteration was improved obviously in the each healing groups. Conclusion: The formula of reforming Shenmaiyin shows promising effect of anti-shock and brain protection, and promoting blood flow and detoxification may be a good Chinese medicine treatment method for mid-anaphase of septic shock.
