摘要
应用微生物转化法,以黑根霉RN-M246为实验菌种,雄甾-4-烯-3,17-二酮为底物,研究了雄甾-4-烯-3,17-二酮的11α-羟基化反应,得到的产品11α-羟基雄甾-4-烯-3,17-二酮是治疗心脑血管疾病的新型药物的重要中间体。实验考察了投料时间等因素对转化生成11α-羟基雄甾-4-烯-3,17-二酮的影响。结果表明,每500mL摇瓶中装入75mL发酵培养基,同时加入底物,接入4%的菌悬液,在28℃发酵72h,转化率可达47.8%,提高了约12%。
The 11α-hydroxylation of androst-4-ene-3,17-dione was catalyzed by Rhizopus nigricans RN-M246 and the product is C11α-hydroxy-androst-4-ene-3,17-dione, which is a key intermediate for a new drug used in the treatment of hypertension and congestive heart failure. The effect of feeding time of raw material on the transformation was investigated. The conversion rate was up to 47.8% under the optimal condition, i.e., culture was performed at 28 ℃ for 72 h in 500 mL shaking flask containing 10 mmol/L substrate and 75 mL media.
