牛大力对^(60)Coγ射线致小鼠造血系统损伤的保护作用 被引量:9

摘要 目的探讨牛大力对60Coγ射线致造血系统损伤的保护作用。方法 40只小鼠分为阴性对照组、辐射模型组、牛大力低[5 g/(kg.d)]、中[10 g/(kg.d)]、高[20 g/(kg.d)]剂量组。除阴性对照组外,用5 Gy的60Coγ射线对各组小鼠进行一次性全身均匀照射以建立辐射损伤模型。牛大力组小鼠在照射前4 d及照射后10 d,以相应剂量的牛大力煎煮液连续灌胃14 d,最后一次灌胃24 h后取血并处死小鼠,检测各组小鼠红细胞数、白细胞数、血小板数、淋巴细胞数,计算脾指数和胸腺指数,通过彗星试验检测小鼠脾、骨髓细胞的DNA损伤。结果与阴性对照组比,辐射模型组、牛大力各剂量组小鼠的红细胞数、白细胞数、血小板数、淋巴细胞数、胸腺指数、脾指数明显减少(P<0.01或P<0.05),脾、骨髓细胞的尾部DNA百分率和尾矩明显增大(P<0.01);但与辐射模型组比,牛大力剂量组小鼠的红细胞数、白细胞数、血小板数、淋巴细胞数、胸腺指数、脾指数明显增加(P<0.01或P<0.05),脾、骨髓细胞的尾部DNA百分率和尾矩明显减少,呈现明显的量效关系。结论牛大力对60Coγ射线致小鼠造血系统损伤具有一定的修复和保护作用,但不能完全拮抗造血系统的损伤。 AIM To study the protective effect of Millettiae speciosae Radix on hemopoietic system of 60Coγ-ray irradiated mice.METHODS Forty mice were divided into negative control group,radiation model group,Millettiae speciosae Radix groups with low [5 g/(kg·d)],middle [10 g/(kg·d)] and high [20 g/(kg·d)] doses.Except in the negative control group,the other mice were irradiated with 5 Gy 60Coγ-rays.Millettiae speciosae Radix groups mice were given with Millettiae speciosae Radix for four days before irradiation and then ten days after irradiation,and negative control group and radiation model group were given with normal saline by gavage once a day.After 24 hours of the last dose,the number of the red blood cells(RBC),white blood cells(WBC),platelets(PLT),lymphocytes(Lymph) and the spleen index and thymus index were detected.DNA damage of bone marrow cells and spleen cells was detected by comet assay.RESULTS Compared with the negative control group,the number of RBC,WBC,PLT,Lymph,spleen index and thymus index of the mice in the radiation model group and Millettiae speciosae Radix groups significantly reduced(P0.01 or P0.05),and tail DNA rate and tail moment of bone marrow cells and spleen cells significantly increased(P0.01 or P0.05).But compared with the radiation model group,the number of RBC,WBC,PLT,Lymph,spleen index and thymus index of the mice in Millettiae speciosae Radix groups significantly increased(P 0.01 or P 0.05),and tail DNA rate and tail moment of bone marrow cells and spleen cells significantly reduced(P0.01 or P0.05) in dose-dependent manner.CONCLUSION Millettiae speciosae Radix has protective effect on mice hematopoietic system damage induced by 60Coγ-rays,but cannot completely antagonize.
