摘要
为了提高丁香叶中总黄酮类化合物的提取率,试验利用超声波辅助提取法,在确定单因素条件的基础上,采用响应曲面设计方法,以总黄酮提取率为响应值,选择溶剂体积、溶剂浓度、提取时间进行3因素3水平优化试验设计,并对结果进行多元回归分析后建立各因素对丁香叶中总黄酮提取率影响的二次多项回归模型。结果表明:最佳工艺条件为液料比20∶1,乙醇浓度55%,提取时间50 min,提取次数2次;在此工艺条件下,丁香叶总黄酮提取率预测值为13.53%,验证值为(13.53±0.13)%。说明建立的优化数学回归模型能较好地预测丁香叶中总黄酮的提取率,优化的工艺条件具有实用参考价值。
To improve the extraction rate of total flavonoids from the leaves of Syzygium aromaticum, an ultrasonic - assisted extraction method was used. Based on the determination of the single factor conditions, the volume of solvent, solvent concentration, extraction time were selected for a three - factor, three'- level central composite design using response surface design method with the extraction rate of total flavonoids as a response value. Based on the effects of all factors on extraction rate of total flavonoids from the leaves of Syzygium aromaticum, a quadratic poly- nomial regression model was established after analyzing the results of multiple regression analysis. The results showed that the optimum extrac- tion condition were obtained as follows : liquid - material ratio of 20:1, ethanol concentration of 55%, extraction time of 50 rain, and two extrac- tion times. Under these conditions, the predicted value of the extraction rate of total flavonoids from the leaves of Syzygium aromaticum was 13.53% , and its verification value was( 13.53± 0.13 ) %. The results indicate that the established model can predict the extraction rate of total flavonoids from the leaves of Syzyglum aromaticurn well, and the optimized extraction conditions have a practical value.
