腺病毒介导睫状神经营养因子基因转染嗅鞘细胞移植治疗颅内段视神经损伤 被引量:1

Effects of olfactory ensheathing cells transfected with ciliary neurotrophic factor mediated by ade-novirus intraocular transfer on intracranial optic nerve injury

摘要 目的研究腺病毒介导睫状神经营养因子(CNTF)转染嗅鞘细胞移植对大鼠颅内段视神经损伤的修复作用。方法构建额下显微外科入路大鼠颅内段视神经损伤动物模型并分组,观察损伤后不同时间点各组大鼠闪光视觉诱发电位(F—VEP)的改变,通过荧光金逆行标记的方法,检测损伤后不同时间点各组大鼠荧光金标记的视网膜神经节细胞(RGC)计数。结果F—VEP检测显示大鼠伤后P1波振幅降低,峰潜时延长(P〈0.05),腺病毒介导CNTF转染嗅鞘细胞移植组较对照组波幅高,峰潜时短(P〈0.01);伤后腺病毒介导CNTF转染嗅鞘细胞移植组视网膜荧光金标记RGC计数较对照组明显增加(P〈0.01)。结论腺病毒介导CNTF转染嗅鞘细胞移植对大鼠颅内段视神经损伤后早期功能的恢复有明显的促进作用。 Objective To investigate the repairment effects of olfactory ensheathing cells (OECs) transfected with ciliary neurotrophic factor(CNTF) mediated by adenovirus intraocular transfer on intracranial optic nerve injury in rats. Methods Intracranial optic nerve injury model of rats were es-tablished by the microsurgery through subfrontal approach and the rats were averagely divided into differ-ent groups. The flash visual evoked potential(F-VEP) of different groups were observed at different time points after injury and the numbers of retinal ganglion cells (RGCs) were retrogradely labeled by fluoro-gold. Results In F-VEP test,the P1 wave amplitude of rats post injury decreased and latency increased (P 〈 0.05). The P1 wave amplitude of olfactory ensheathing cells transfected with CNTF mediated by adenovirus group was higher than that in the control group and the latency was shorter meantime, there was significant difference(P 〈0.01 ). The numbers of labeled retinal ganglion cells of olfactory ensheat-hing cells transfected with CNTF mediated by adenovirus group was significantly higher than that in the control group ( P 〈 0. 01 ). Conclusions The transfer of olfactory ensheathing cells transfected with CNTF mediated by adenovirus intraocular on rats after intracranial optic nerve injury can improve the re-pairment of visual function during early period.
