摘要
目的 :探讨人参皂甙抗缺血缺氧性脑损伤作用是否与抑制谷氨酸的释放有关 ,为临床应用人参皂甙防治缺血缺氧性脑损伤提供理论依据。方法 :超微观察大鼠离体海马脑片缺氧时 CA1区锥体细胞结构及含谷氨酸的突触小泡 ;并在培养的小鼠皮质神经细胞观察模拟缺血时谷氨酸的释放。同时 ,观察人参皂甙对上述谷氨酸释放的影响。结果 :缺氧时海马 CA1区含谷氨酸的突触小泡明显减少甚至耗竭 ,培养的小鼠皮质神经细胞缺血时谷氨酸释放明显增加。使用人参皂甙对上述两种形式谷氨酸的释放均有明显抑制作用。结论 :人参皂甙抗缺血缺氧性脑损伤确实与抑制脑内谷氨酸释放有重要的关系。
Objective: To investigate whether the antagonism of ginsenosides against hypoxic-ischemic brain damage is related to the inhibition of glutamate release, and afford a theoretical basis of clinical administration of ginsenosides for treatment of hypoxic-ischemic brain damage.Methods: We observed the CA1 pyramidal neurons' structure and glutamate synaptic vesicle numbers of the rat hippocampal slice with electronmicroscope, and glutamate release from the cultured cortical neurons of mouse during hypoxia and simulated ischemia respectively. In the same time, we observed the influences of ginsenosides on these two types of glutamate release.Results: The glutamate synaptic vesicle numbers decreased and even exhausted, and the glutamate release from the cultured cortical neurons increased markedly. After the administration of ginsenosides,the above two types of glutamate release were inhibited.Conclusion: The antagonist effect of ginsenosides against hypoxic-ischemic brain damage is indeed related to the inhibition of glutamate release.
