摘要
目的:研究原花青素B2对人表皮细胞株HaCaT增殖功能的影响。方法:常规培养HaCaT细胞,将细胞分为对照,6.25、12.5、25、50、100μmol/L原花青素B2处理组及阳性对照rhEGF处理组,采用MTT法检测各组细胞在24h和48h增殖水平。然后将细胞分为对照组,25、50μmol/L原花青素B2处理组和阳性对照rhEGF处理组,采用细胞划痕实验检测细胞各组细胞划痕愈合率。结果:培养24h和48h后,12.5、25、50、100μmol/L细胞相对增殖率均明显高于对照组(P<0.05),且具有一定的浓度依赖性。6.25μmol/L原花青素B2处理组与对照组无明显差异。培养24h后,25、50μmol/L原花青素B2组划痕愈合率分别为41.7%、40.2%,显著高于对照组的28.1%(P<0.05)。培养48h后,各实验组划痕愈合率均为100%,显著高于对照组78.9%(P<0.05),但两实验组之间并无明显差异。结论:原花青素B2能够促进HaCaT细胞增殖和细胞划痕损伤后愈合,有望成为治疗和预防皮肤老化和创伤的有效药物。
Objective: To study the influence of proanthocyanidin B2 on the proliferation of human epidermal cell line HaCaT. Methods: HaCaT cells were routinely cultured and divided into control group , 6.25, 12.5, 25, 50, 100μmol/L proanthocyanidin B2 treated groups and rhEGF treated group. Cells were cultured with proanthocyani- din B2 for 24h or 48h. MTT assay was used to test the influence of proanthocyanidin B2 on the cell proliferation. Then the cells were divided into control, 25μmol/L, 50μmol/I. proanthocyanidin B2 treated groups and rhEGF trea- ted group. Cell scratch assay was conducted in each group. The pictures of cell were taken at post scratch hour 0, 24 and 48, and the wound area healing rate was calculated. Results: After 24h and 48h treatment, the relative growth rate in 12.5, 25, 50, 100μmol/L proanthocyanidin B2 treated groups were significantly higher than control (P〈0. 05). The 6.25/μM proanthocyanidin B2 treated groups showed no difference with control. The wound area healing rate in 25μmol/L (41.7%) and 50μM (40.2%) groups at 24 hour was significantly higher than that in con- trol group (28. 1%,P〈0.05). At 48 hour, the wound area healing rate in 25μmol/L (100%) and 50μmol/L (100%) groups was significantly higher than that in control group (78.9%, P〈0.05). But there was not signifi cantly different beween 25μmol/L and 50μmol/L groups. Conclusion: Proanthocyanidin B2 could promote prolifera- tion of HaCaT cell. The encouraging in vitro results should be taken into consideration when evaluating the clinical pharmacology of plant--derived proanthocyanidin B2 as a novel agent for skin would healing.
