旋覆花素对阿尔茨海默病模型大鼠脑海马组织Bax,Bcl-2表达的影响 被引量:3

摘要 目的观察旋覆花素对Aβ25-35海马内注射所致阿尔茨海默病(AD)模型大鼠海马Bax,Bcl-2表达的影响。方法30只sD大鼠随机分为对照组、模型组及旋覆花素给药组,Aβ25-35海马内注射制备AD大鼠模型,给药组给予旋覆花素26mg·kg^-1·d^-1、模型组给予等量溶剂,分两次灌胃,共21d。免疫组化及Western印迹测定大鼠海马Bax,Bcl-2表达。结果模型组海马Bax表达升高,给药组Bax表达下降(P〈0.05)。对照组大鼠脑海马组织Bcl-2表达量与模型组及旋覆花素给药组大鼠脑海马组织表达量无明显差别(P〉0.05)。结论旋覆花素主要通过抑制促凋亡基因Bax的表达,从而抑制Aβ诱导的AD模型大鼠脑海马的神经细胞凋亡,而对凋亡抑制基因Bcl-2表达影响不大。 Objective To explore the effect of inulicin on expressions of Bax, Bcl-2 in hippocampus of AD rats. Methods SD rats were microinjected with Aβ25 -35 into hippocampus to induce AD model rats. Rats in inulicin treatment group were pretreated with inuliein (contain ABL 2. 66 mg/ml) dissolved in tween-80 3 days before operation, then inulicin was administered to rats at dose of ABL 26 mg·kg^-1·d^-1 by gastric gavage orally for 18 days (n = 10). The Bcl-2 and Bax protein in hippoeampus were detected by immunohistoehemistry and western blot. Results Bax expression was more intensive in hippoeampus cell of model rat than that of control rat ( P 〈 0.05 ), but there was no difference in inulicin-treated and control group ( P〉0. 05 ). There were no difference in Bcl-2 expression among model, innuli- ein-treated and control rats ( P 〉 0. 05 ). Conclusions Inuliein could inhibit the apoptosis by Aβ injection into hippocampus by down-regu- lating Bax expression, but not up-regulating Bcl-2 expression.
