摘要
目的:探讨慢性阻塞性肺疾病(COPD)患者肺组织和血清中高迁移率族蛋白B1(HMGB1)的表达,及与TNF-α和IL-1β的相关性。方法:RT-PCR及免疫组织化学染色法检测24例肺叶切除术患者(COPD组和对照组各12例)肺组织中HMGB1的mRNA和蛋白表达。ELISA法检测同期COPD患者AECOPD组78例,COPD稳定期组40例及健康者40例血清中HMGB1、TNF-α和IL-1β水平。结果:COPD组肺组织中HMGB1平均光密度值和mRNA相对转录水平均显著高于对照组。AECOPD组和COPD稳定期组血清中HMGB1、TNF-α和IL-1β水平均显著高于对照组。AECOPD组血清中HMGB1与TNF-α和IL-1β水平均呈正相关。结论:HMGB1在COPD患者肺组织和血清中表达水平显著升高,提示可能参与COPD的炎症反应过程。HMGB1血清水平测定有助于反映COPD患者病情的严重程度。
Objective To explore the expressions of high mobility group protein box-1 (HMGB1) in lung tissue and serum of patients with chronic obstructive pulmonary disease (COPD) and to explore its relationship with TNF-α and IL-1β. Methods The lung tissues were obtained from 12 patients with COPD and 12 patients without COPD, and then the expressions of HMGB1 mRNA and protein were detected by RT-PCR and immunohistochemieal staining, respectively. Blood samples were taken from 78 patients with AECOPD, 40 outpatients with stable COPD and 40 healthy volunteers. Serum concentrations of HMGB1, TNF-α and IL-1β were detected by ELISA. Results The average optical density and the mRNA relative transcription level of HMGB1 in COPD group were significantly higher than those in normal lung tissue control group. The serum concentrations of HMGB1, TNF-α and IL-1β in patients with AECOPD and in patients with stable COPD were significantly higher than those in healthy controls. HMGB1 were correlated positively with TNF-α and IL-1β in AECOPD group. Conclusions The over expression of HMGB1 may play an important role in the inflammatory response of COPD. The measurement of serum HMGB1 was useful in evaluating the severity of COPD.
