摘要
目的 :建立血清中茶碱的提取和薄层色谱扫描定性和定量检测方法。方法 :血清用 1%盐酸酸化 ,乙醚 二氯甲烷 (6∶4)或氯仿萃取 ,点于GF2 54 薄层板上 ,正庚烷∶三氯甲烷∶无水乙醇∶乙酸乙酯∶甲醇∶冰乙酸 (5∶2∶2∶1.5∶2 .5∶0 .5 )展开 ;薄层扫描检测。结果 :茶碱薄层层析Rf值为 0 .35 ;最大吸收波长为 2 75nm ;血中茶碱线性检测范围为 1~ 2 4μg/ml,最小检出浓度为 0 .4μg/ml。 结论 :本法快速、简便、定性定量准确 ,可用于茶碱中毒的快速诊断和体内药物浓度监测。
Objective::To establish a rapid method for determination of theophylline in serum.Methods:Serum sample is acidified with 1% HCl,then extracted with ether dicholromethane(v/v= 6/4) or chloroform.The extract is applied to a GF 254 TLC plate and developed with a solvent mixture of n heptane∶chloroform∶ethanol∶ethyl acetate∶methanol∶acetic acid(5∶2∶2∶1.5∶2.5∶0.5) and then determined by TLC scanning.Results:The Rf value for theophylline is 0.35 with a maximum absorption at 275 nm.The liner range of theophylline is 1~24μg/ml with a detection limit of 0.4μg/ml.Conclusion:A rapid,convinient,and accurate method for qualitative and quantitative determination of theophylline in serum is established.Theophylline poisoning and serum theophylline level can be determined by this method.
