摘要
采用直接法和间接法进行了99mTc标记重组水蛭素的研究,结果表明,间接法制备的99mTc-重组水蛙素具有更高的放化纯和稳定性。更适合于血栓模型的显像研究。99mTc-重组水蛙素在小鼠体内的分布表明,心、肝、脾、肺和脑的放射性分布均较低;放射性主要集中在肾脏,15min时肾脏放射性可达总注入剂量的65%;且血液中放射性清除较快。兔血栓模型显像结果表明,99mTc-重组水蛙素可在血栓部位浓聚;注入后1h即可使血栓清晰显影。因此,99mTc-重组水蛙素有望成为一种新型的血栓显像剂。
Comparsion of two labeling methods showed that 99mTc-recombinant hirudin (99mTc-rH) prepared by modified 2-imiothiolane method had higher radiochemical purity and stability than that by direct labeling method. Biodistribution of 99mTc-rH in mice showed that the radioactivity in heart, lung, liver and spleen was very low, while that in kidney was high. 99mTc-rH was cleared rapidly in blood and excreted mostly from kidney. The arterial and venous thrombi in rabbits were clearly imaged 60min after injection of 99mTc-rH, T/B ratio was 2.24 and 1.95 respectively. The rH would be a potential agent for diagnosis of thrombotic disease.
