摘要
本文用 15种识别 6碱基的限制性内切酶ApaⅠ、BamHⅠ、BglⅠ、BglⅡ、ClaⅠ、DraⅠ、EcoRⅠ、EcoRⅤ、HaeⅡ、HindⅢ、KpnⅠ、PvuⅡ、PstⅠ、SacⅠ和SalⅠ对绵羊、山羊和岩羊mt NDA的限制性片断长度多态性进行了比较研究 ,以探讨其遗传分化关系。共检测到 75个酶切位点 ,4 1种限制性态型 ,其中绵羊和山羊、绵羊和岩羊以及山羊和岩羊的共享位点数分别为 15、8和14 ,共享位点所占比率分别为 19 74 %、10 53%和 18 42 %。绵羊和山羊、绵羊和岩羊以及山羊和岩羊基本单倍型间的遗传距离分别为 0 0 557、0 0 6 4 0和 0 0 311,平均mtDNA多态度 (或称核苷酸差异均数 )分别为 5 7182 %、6 584 7%和 3 186 0 %
WT5BZ]:The restriction fragment length polymorphism(RFLP) of mitochondrial DNA (mtDNA) of sheep,goat and bharal were studied using 15 restriction endonucleases recognizing 6 base pair,Apa Ⅰ,BamH Ⅰ,Bgl Ⅰ,Bgl Ⅱ,Cla Ⅰ、Dra Ⅰ,EcoR Ⅰ,EcoR Ⅴ,Hae Ⅱ,Hind Ⅲ, Kpn Ⅰ, Pst Ⅰ, Pvu Ⅱ, Sac Ⅰ and Sal Ⅰ,in order to investigate the phylogenetic relationship between them The results indicated that 76 restriction sites and 40 restriction morphs were detected,which could be sorted into 9 haplotypes Between sheep and goat,sheep and bharal,goat and bharal,the fragments shared together by them were 15,8 and 14 respectively,the ratios of fragments shared together were 19 74%,10 53%and 18 42%respectively,the genetic distance were 0 0557,0 0640 and 0 0311, the average polymorphic degree (π value) were 5 7182%,6 5847% and 3 1860%
