摘要
目的通过大鼠孕期不同阶段染铅制备仔鼠染铅模型,测定仔鼠血铅及脑铅水平,并观察S100B蛋白在不同染铅模型仔鼠脑组织中的表达及意义。方法将72只健康清洁级Wistar妊娠大鼠随机分为孕早期染铅组(孕早期:妊娠1~10d饮用0.25g/L醋酸铅溶液;孕后期:妊娠11~20d饮用蒸馏水)、孕晚期染铅组(孕早期饮用蒸馏水,孕后期饮用0.25g/L醋酸铅溶液)、孕全程染铅组(妊娠1~20d饮用0.25g/L醋酸铅溶液)和对照组(孕全程饮用蒸馏水),每组18只。各组妊娠末期剖宫取仔鼠,采血测定血铅水平;完整取出仔鼠大脑,测定脑铅水平和S100B蛋白的表达。结果仔鼠血铅及脑铅水平由低至高依次为对照组、孕晚期染铅组、孕早期染铅组、孕全程染铅组,任意两组比较,差异均有统计学意义(F=12.01、7.39,P<0.05)。与对照组比较,孕期染铅各组子代仔鼠脑组织中S100B蛋白的表达均升高;与孕全程染铅组比较,孕早期及孕晚期染铅组子代仔鼠脑组织中S100B蛋白的表达均下降,差异有统计学意义(F=3.73,P<0.05)。结论孕期铅暴露使得子代仔鼠脑组织中S100B蛋白的表达升高,以孕全期铅暴露组表达水平最高。母孕期低水平铅暴露导致子代仔鼠脑组织中S100B蛋白表达异常可能是铅致脑损伤的毒性机制之一。
Objective A newborn rat model with lead exposure was created through lead exposure to a gravid rat during different gestation period. The blood and brain lead levels in newborn rats were detected and the expression of S100B protein in brain tissue of lead exposure models determined. Methods A total of 72 pregnant Wistar rats were equally randomized to four groups as early gestation lead-exposure group: 0.25 g/L lead acetate was given orally on days 1-10 of gestation, and distilled wa ter on days 11--20; late gestation lead-exposure group: oral distilled water in early stage, and 0.25 g/L lead acetate in late stage; whole gestation lead exposure group: oral 0.25 g/L lead acetate in the whole gestation; control group: oral distilled water in the whole gestation. New-born rats were obtained surgically and blood lead levels detected. The brains were completely removed for de- tection of S100B protein expression. Results The levels of blood lead and brain lead, from low to high, were in this order: con- trol group, late-gestation group, early gestation group and whole gestation group, A comparison between any two groups (with re- gard to the blood lead and brain lead level) showed significant difference (F: 12.01,7.39 ;P〈0.05). Compared with the control, the expressions of S100B in brain tissues of lead-exposed offspring rats were significantly higher; Compared with the whole gesta- tion group, the expressions of S100B in early and late gestation group were lower (F=3.73,P〈0.05). Conclusion Lead expo- sure during gestation results in elevation of S100B expression in brain tissue of offspring rats, the highest expressions are observed in those of whole phase of lead exposure. Low lead exposure during mother's pregnancy induces abnormal S100B expression in brain tissue of offspring rats is likely to be one of the toxic mechanisms of lead-induced brain injury.
