摘要
通过固体培养基平板添加除草剂精喹禾灵进行微拟球藻高脂藻株筛选,发现7.5μmol/L精喹禾灵可抑制99.7%的微拟球藻生长,10μmol/L精喹禾灵完全抑制微拟球藻生长。筛选出2株抗7.5μmol/L精喹禾灵藻株KA1和KA2。培养52d后,其总脂含量达干重的46.73%和38.60%,分别比出发藻株提高了29.02%和6.57%。筛选藻株KA1的比生长速率和终生物量与出发藻株无显著差异,但其油脂产率比出发藻株显著提高。确定了用精喹禾灵筛选高脂微藻的有效性,可为筛选高脂藻株、推进微藻生物柴油开发提供参考。
The lipid-rich Nannochloropsis oceanica was screened out by adding the herbicide quizalofop-p-ethyl to the solid medium of the microalga.It was found that 7.5mmol/L of quizalofop-p-ethyl(final concentration in medium) inhibited the growth of 99.7% of N.oceanica cells and 10mmol/L of quizalofop-p-ethyl inhibited the growth of N.oceanica completely.Two quizalofop-p-ethyl resistant strains of N.oceanica,namely KA1 and KA2,were obtained on f/2 medium containing 7.5mmol/L quizalofop-p-ethyl.When cultured for 52 days,the total lipid of KA1 andKA2 increased by 29.02% and 6.57% over that of the original strain,reaching 46.73% and 38.60% of dry weights,respectively.The specific growth rate and the final dry biomass of KA1 were not significantly different from the original strain;however the oil productivity of KA1 was higher than that of the original.The applicability of herbicide quizalofop-p-ethyl to the screening of lipid-rich microalgae was evidenced in this study,which is promising for screening lipid rich microalgae and advancing microalgal diesel production.
