摘要
目的探讨鼻咽癌中胰岛素样生长因子1受体(insulin-like growth factor 1 receptor,IGF-1R)、CD147、Fascin的表达和相关性,及其与鼻咽癌生物学行为的关系。方法采用免疫组化SP法检测50例非角化性未分化型鼻咽癌原发灶(其中无颈部淋巴结转移9例、颈部淋巴结转移41例)和30例鼻咽黏膜慢性炎中IGF-1R、CD147、Fascin蛋白的表达。结果鼻咽癌细胞中IGF-1R、CD147和Fascin蛋白的阳性率均高于鼻咽黏膜柱状上皮细胞,组间差异均有统计学意义(P<0.01)。鼻咽癌中IGF-1R与CD147蛋白表达呈正相关(P<0.05);IGF-1R与Fascin蛋白表达呈正相关(P<0.05);CD147与Fascin蛋白表达呈显著正相关(P<0.01)。颈部淋巴结转移组中IGF-1R、CD147、Fascin蛋白的阳性率均明显高于无颈部淋巴结转移组(P<0.01)。3种蛋白在Ⅲ、Ⅳ期组鼻咽癌细胞中阳性率均高于Ⅱ期组(P<0.05)。结论 IGF-1R、CD147、Fascin蛋白在鼻咽癌中均呈高表达,三者表达呈正相关,可能共同参与鼻咽癌的侵袭、转移过程,有望成为鼻咽癌进展的潜在标志物。
Purpose To investigate the expression of insulin-like growth factor 1 receptor ( IGF-1R), CD147 and Fascin protein in nasopharyngeal carcinoma (NPC) and to find out their correlations with the biologic behaviors of the tumor. Methods Immunohisto-chemistry SP method was adopted to detect the protein expression of IGF-1 R, CD147 and Fascin in 30 cases of chronic nasopharyngitis and 50 cases of non-keratinizing undifferentiated NPC (9 non-metastatic and 41 metastatic cases). Results Positive staining rates of IGF-1 R, CD147 and Fascin protein in 50 cases NPC were higher than those of nasopharyngeal mucosal columnar epithelia (P 〈 0. 01 ) . There was statistically positive correlation between expression of IGF-1R and CD147 protein ( P 〈 0. 05 ), between positive expression of IGF-1R and Fascin protein ( P 〈 0. 05 ), and between expression of CD147 and Fascin protein ( P 〈 0. 01 ). Positive rates of IGF- 1R, CD147 and Fascin protein in cervical lymph node metastasis groups were markedly higher than those of cervical lymph node no-metastasis ones (P 〈0. 01 ). It also showed positive expression of three proteins in stage m and 1V groups was higher than those in stage II group (P 〈 0. 05). Conclusions There is a high level of expression of IGF-1 R, CD147 and Fascin proteins in NPC, in which the three proteins are positively related to the invasion and metastasis of NPC. IGF-1R, CD147 and Fascin proteins might be po- tential candidates of biological biomarkers for progressing NPC.
