接受高效抗逆转录病毒治疗的艾滋病患者γδT细胞表型和功能分析 被引量:2

Phenotype and functional analysis of γδ T cells in AIDS patients with highly active antiretroviral therapy

摘要 目的研究接受高效抗逆转录病毒治疗(highly active antiretroviral therapy,HAART)的艾滋病(acquired immunodeficiency syndrome,AIDS)患者外周静脉血中γδT细胞表型和功能的变化。方法接受HAART治疗的患者(n=36)来自首都医科大学附属北京佑安医院2005-2012年接受免费抗病毒治疗队列,健康对照者(n=17)来源于实验室招募的健康志愿者。分离两组研究对象外周血单个核细胞(pe-ripheral blood mononuclear cells,PBMCs)后,采用流式细胞表面染色法检测外周静脉血中γδT细胞及其亚型的比例和γδT细胞表面活化分子(CD38和HLA-DR)的表达;采用流式细胞内染色法(intracellularstaining,ICS)检测γδT细胞干扰素(interferon,IFN)-γ和白细胞介素(interleukin,IL)-17的分泌。结果与健康对照者(healthy controls,HCs)比较,接受HAART治疗患者外周血Vδ2+γδT细胞的比例显著下降[(18.33%±20.20%)vs(59.54%±30.87%),P<0.01],而Vδ1+γδT细胞的比例明显增加[(38.35%±23.99%)vs(16.63%±17.69%),P<0.01]。接受HAART治疗的患者γδT细胞表面CD38和HLA-DR分子的表达显著增加(P<0.01)。然而,与健康对照者比较,接受HAART治疗患者分泌IFN-γ和分泌IL-17的γδT细胞的数量差异无统计学意义。结论单纯HAART治疗既不能增加Vδ2+γδT细胞的数量和γδT细胞细胞因子分泌的能力,也不能降低艾滋病患者γδT细胞的活化水平。 Objective To analyze the phenotype and function of peripheral blood 78 T cells in acquired immuno- deficiency syndrome (AIDS) patients receiving highly active antiretroviral therapy (HAART). Methods Pe- ripheral blood mononuclear cells (PBMCs) were isolated from AIDS patients and healthy controls, γδ T cells, subset of γδ T cells, and the active markers of CD38 and HLA-DR expressed on 78 T cells were examined by flow cytometry. Secretion of interferon gamma (IFN-γ) and interleukin 17 (IL-17) was detected by intracel- lular staining (ICS). Results A total of 36 patients were included from a cohort of AIDS patients receiving free HAART therapy, and 17 healthy controls were recruited from volunteers enrolled in our laboratory. Thefrequency of V32+ 73 γδ cells in the HAART treatment group is significantly lower than that in healthy controlsE ( 18.33 % ±20.20 % ) vs (59.54% ± 30.87 % ), P〈0.011, while the frequency of Vδ1 + γδ T cells is signif- icantly higher than that in the control group[- (38.35%±23.99%) vs (16.63%±17.69%), P〈0.011. The expression of CD38 and HLADR on γδ T cells is markedly elevated in the treatment group than that in the control group (P〈0.01). However, there were no significant differences in the number of γδ T cells secreting IFN-γand ILl7 between these two groups. Conclusions HAART treatment only does not increase the number af V^2+ γδ T cells ar reduce the activitv of γδ T cells in AIDS natients.
