摘要
目的研究壳寡糖对紫外线(UVB)照射成纤维细胞增殖及基质金属蛋白酶(MMP)表达的影响。方法以原代培养的成纤维细胞为研究对象,用UVB紫外照射仪照射细胞后培养,通过逆转录聚合酶链反应(RT-PCR)、细胞免疫荧光、酶联免疫吸附测定(ELISA)法等检测细胞增殖和MMP表达情况。结果原代培养成纤维细胞经UVB辐射后,细胞增殖活性低于未辐射的正常对照组(P<0.05),壳寡糖处理细胞组与单纯UVB照射组比较,细胞增殖活性增强,差异有统计学意义;壳寡糖处理细胞组MMP-1及MMP-3 mRNA的表达较单纯UVB照射组下降,MMP-1及MMP-3蛋白的表达亦明显下降。结论壳寡糖对紫外照射成纤维细胞有保护作用,壳寡糖可抑制MMP-1及MMP-3 mRNA及蛋白高表达。
Objective To study effect of chitosan oligosaccharide on proliferation of human dermal fibroblasts exposed to ultraviolet B (UVB) and the expression of matrix metalloproteinase (MMP)-I and MMP-3. Methods Primary fibroblasts were cultivated and exposed to UVB. Cell proliferation and MMP expression were determined by reverse transcription polymerase chain reaction (RT-PCR), immunocytofluoreseence and ELISA. Results Proliferation of the fibroblasts being exposed to UVB was significantly inhibited compared with that of the control without UVB exposure and ehitosan oligosaccharide treated group ( P〈 O. 05 ). MMP-1 and MMP-3 mRNA expression and protein levels in chitosan oligosaccharide treatment group were down-regulated in comparison to those in the untreated UVB exposure group. Conclusion Chitosan oligosaccharide protects fibroblasts against UVB exposure,and inhibits mRNA and protein expression of MMP-1 and MMP-3.
