二苯乙烯苷预适应对脑缺血-再灌注损伤大鼠细胞凋亡的保护作用 被引量:2

Protective Effects of Stilbene Glucoside Preconditioning Against Cell Apoptosis in Rats Subjected to Cerebral Ischemia-Reperfusion

摘要 目的探讨二苯乙烯苷(2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside,C20H22O9,TSG)对脑缺血-再灌注损伤大鼠细胞凋亡的保护作用及其机制。方法将48只大鼠按随机数字表法分为6组:正常对照组、假手术组、脑缺血-再灌注损伤组(I/R组)、TSG 1组(TSG 0.038g.kg-1组)、TSG 2组(TSG 0.114g.kg-1组)、TSG 3组(TSG 0.342g.kg-1组),每组8只。TSG 1组、TSG 2组、TSG 3组分别给予TSG 0.038、0.114、0.342g.kg-1,连续灌胃7d;正常对照组、假手术组、I/R组分别给予同体积生理盐水,连续灌胃7d。第8天,正常对照组不作任何处理,断颈处死大鼠;I/R组、TSG 1组、TSG 2组、TSG 3组通过大脑中动脉栓线阻断法建立脑缺血-再灌注模型,再灌注24h后,断颈处死大鼠;假手术组仅进行麻醉和血管分离术,不结扎血管及导入栓线。然后,取6组脑组织进行细胞凋亡、MDA含量及GSH-Px、SOD活性的检测。结果 I/R组的脑组织中凋亡细胞率明显高于假手术组(P<0.05),TSG 1组、TSG 2组、TSG 3组的脑组织中凋亡细胞率均明显低于I/R组(均P<0.05)。I/R组的脑组织中MDA含量明显高于假手术组(P<0.05),SOD、GSH-Px活性均明显低于假手术组(均P<0.05);TSG 1组、TSG 2组、TSG 3组的脑组织中MDA含量均明显低于I/R组(均P<0.05),SOD、GSH-Px均明显高于I/R组(均P<0.05)。结论 TSG预处理对脑缺血-再灌注损伤大鼠细胞凋亡有显著的保护作用,其机制与增强内源性抗氧化剂活性有关。 Objective To investigate the protective effects of stilbene glucoside(2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside,C20H22O9,TSG) against cell apoptosis and the underlying mechanism in rats subjected to cerebral ischemia-reperfusion(I/R) injury.Methods Forty-eight rats were randomly divided into six groups.Rats in TSG 1,TSG 2 and TSG 3 groups were intragastrically given TSG 0.038,0.114 and 0.342 g·kg-1 for 7 days,respectively.Rats in control group,sham operation group and I/R group were given the same volume of normal saline.On the eighth day,rats in control group were executed by decapitation.Rats in I/R,TSG 1,TSG 2 and TSG 3 groups were sacrificed at 24 hours after reperfusion following suture middle cerebral artery occlusion.Rats in sham operation group only received anaesthesia and vascular dissection,without suture ligation and thread insertion.Then,brains were collected for the detection of cell apoptosis,MDA content and GSH-Px and SOD activity.Results Compared with I/R group,apoptosis rate and MDA content significantly decreased but GSH-Px and SOD activity significantly increased in sham operation,TSG 1,TSG 2 and TSG 3 groups(P〈0.05).Conclusion TSG preconditioning can protect cells against I/R injury-induced apoptosis through enhancing endogenous antioxidant activity in rats.
