何首乌提取物对大鼠脑缺血再灌注损伤的保护作用 被引量:12

摘要 目的:观察何首乌提取物对大鼠局灶性脑缺血再灌注后超氧化物歧化酶活性、丙二醛及一氧化氮含量的影响,探讨其对脑损伤的保护作用。方法:实验于2003-04/05在山东大学医学院实验室完成。40只Wistar雄性大鼠随机分为4组:假手术组、缺血组、20mg/kg何首乌组和40mg/kg何首乌组,每组10只。制造大鼠大脑中动脉缺血再灌注模型。20mg/kg何首乌组和40mg/kg何首乌组于缺血前30min及缺血后1h分别给予何首乌提取物20mg/kg、40mg/kg腹腔内注射。各组大鼠于再灌注后24h检测脑缺血组织超氧化物歧化酶活性、丙二醛和一氧化氮含量。结果:40只大鼠均进入结果分析。①超氧化物歧化酶活性:20mg/kg何首乌组、40mg/kg何首乌组明显高于缺血组[(85.1±10.2),(103.2±12.9),(62.8±8.7)NU/mg,P<0.01],高剂量组高于低剂量组(P<0.05)。②丙二醛和一氧化氮含量:20mg/kg何首乌组、40mg/kg何首乌组明显低于缺血组[丙二醛含量:(6.58±0.62),(5.41±0.58),(9.24±0.88)μmol/g,P<0.01;一氧化氮含量:(5.83±0.77),(4.71±0.59),(7.65±0.86)mmol/g,P<0.01],高剂量组高于低剂量组(P<0.05)。结论:①何首乌提取物可抑制脑缺血再灌注损伤后超氧化物歧化酶活性的下降及丙二醛、一氧化氮含量的升高,表明何首乌提取物可以清除体内过多的氧自由基,对脑缺血再灌注损伤起保护作用。②何首乌提取物的疗效与剂量有关,高剂量的疗效明显好于低剂量。 AIM: To observe the effects of tuber fleeceflower root (TFR) extract on activity of superoxide dismutase (SOD), contents of malondialdehyde (MDA) and nitrogen monoxide in rats after focal cerebral ischemical reperfusion, and explore its protective effects on cerebral injury. METHODS: The experiment was conducted in Experimental Animal Center of Medical College, Shandong University from April to May 2003. Forty Wistar male rats were randomly divided into 4 groups: Shamoperated group, ischemia group, 20 mg/kg TFR group and 40 mg/kg TFR group with 10 rats in each group. The ischemia/reperfusion models of middle cerebral 'artery in rats were made. At 30 minutes before ischemia and 1 hour after ischemia the rats in the 20 mg/kg TFR group and 40 mg/kg TFR group were treated with intraperitoneal injection with 20 mg/kg, 40 mg/kg TFR extract, respectively. At 24 hours after reperfusion, the rats in every group were detected on the activity of SOD, and the contents of MDA and nitrogen monoxide. RESULTS: Forty rats were all involved in the result analysis.① The activity of SOD: It was significantly higher in the 20 mg/kg TFR group and the 40 mg/kg TFR group than that in the ischemia group [(85.1±10.2), (103.2±12.9), (62.8±8.7) NU/mg, P 〈 0.01], and higher the high-dose group than that in the low-dose group ( P 〈 0.05 ). ② Contents of MDA and nitrogen monoxide: It was lower in the 20 mg/kg TFR group and 40 mg/kg TFR group than those in the isehemia group [content of MDA: (6.58±0.62), (5.41±0.58), (9.24±0.88)μmol/g, P 〈 0.01; content of nitrogen monoxide: (5.83±0.77), (4.71±0.59), (7.65±0.86) mmol/g, P 〈 0.01], and higher in the high-dose group than that in the low-dose group ( P〈0.05 ). CONCLUSION: ① TFR extract can inhibit the decrease of SOD activity and the increase of content of nitrogen monoxide after cerebral ischemical reperfusion injury, which indicates that the TFR extract can clear the more oxygen-derived free rad
