摘要
为研究布莱凯特黑牛PDSS1(decaprenyl diphosphate synthase subunit 1)基因与肉质性状的相关性,采用PCR-SSCP技术对96头布莱凯特黑牛进行单核苷酸多态性检测。经PCR-SSCP检测及克隆测序发现,布莱凯特黑牛PDSS1基因各扩增片段中,P4表现为AA、AB、BB 3种基因型,P9表现为CC、CD 2种基因型,且各存在1个SNP位点:第4个外显子122bp处发生A→G的碱基突变,并引起相应氨基酸的改变(N156S);第11个外显子6bp处发生C→T的碱基突变,但未引起氨基酸的改变。经肉质性状关联性分析发现,AA、AB、BB不同基因型间差异不显著(P>0.05),但AB型的综合肉质指标较好。研究表明,布莱凯特黑牛PDSS1基因各外显子核苷酸序列的稳定性极高;外显子4内的碱基突变可能会引起PDSS1蛋白质空间结构及功能的改变,但与肉质性状相关性不显著。
In this study, to provide fundamental basis for studying the correlations between PDSS1 gene and meat quality traits of Black Cattle, a total of 96 animals were selected and the single nucleotide polymorphisms(SNPs) of the exon of decaprenyl diphosphate synthase subunit 1 (PDSS1) gene were detected by PCR-SSCP. The results showed that P4 represented three kinds of genotypes, AA, AB, and BB, while P9 represented two kinds of genotypes, CC and CD, each possessing one SNP loci of A122G in exon 4 or C11T in exon 11 of PDSS1 gene,respectively. The difference of meat quality traits among genotypes AA, AB, and BB was not significant(P〉0.05), but the multiple meat quality indexes of AB type was better. This suggests that the exon sequence of Black Cattle PDSS1 is very stable and the base mutation in exon 4 may cause the change in structure or function of PDSS1 protein but is not correlated significantly to meat quality trait.
