摘要
目的观察罗勒提取物对小鼠腹腔巨噬细胞(RAW264.7)生长及其胞内5-脂氧酶活性的影响。方法采用MTT比色法检测罗勒乙酸乙酯提取物或罗勒正丁醇提取物对正常RAW264.7细胞和经脂多糖刺激的RAW264.7细胞生长的影响,求出其IC50值;用罗勒乙酸乙酯提取物或罗勒正丁醇提取物分别干预经脂多糖处理的RAW264.7细胞,采用ELISA法测定并比较上清液中白三烯B4的质量浓度,求出其抑制率。结果罗勒乙酸乙酯提取物和罗勒正丁醇提取物对正常RAW264.7细胞的生长有不同程度的抑制作用,给药时间为24 h的IC50值分别为312.5μg/mL、350.9μg/mL,具有浓度依赖性;两种提取物对经脂多糖刺激的RAW264.7的生长有一定的增殖作用(P<0.05);罗勒乙酸乙酯提取物和罗勒正丁醇提取物对白三烯B4的最大抑制率分别为42.78%、38.63%,其中罗勒乙酸乙酯提取物的作用强于罗勒正丁醇提取物(P<0.05)。结论罗勒提取物体外对经脂多糖刺激的RAW264.7细胞生长的最大抑制质量浓度为100μg/mL,对细胞内5-脂氧酶活性具有抑制作用。
AIM To study the effects of extracts of Ocimum basilicum L.on mouse peritoneal macrophages RAW264.7 and 5-lipoxygenase pathway.METHODS Effects of Ocimum basilicum L.extracts on growth of normal RAW264.7 cells and lipopolysaccharide-stimulated RAW264.7 cells by MTT colorimetric assay and the IC50 values were calculated to screen out the dose for administration.The lipopolysaccharide-induced RAW264.7 cells were intervened with Ocimum basilicum L.ethyl acetate extract and Ocimum basilicum L.butanol extract respectively.The concentrations of leukotriene B4 in the supernatant were measured by ELISA approach and the inhibition rate was calculated.RESULTS Ocimum basilicum L.ethyl acetate extract and Ocimum basilicum L.butanol extract inhibited on the growth of normal RAW264.7 cells,in which IC50 in 24 h were 312.5 μg/mL and 350.9 μg/mL respectively with concentration-dependent model.Both extracts had certain proliferation on growth of lipopolysaccharide-stimulated RAW264.7 cells(P0.05).The maximal inhibiting ratio were 42.78% and 38.63% respectively,and the inhibiting ratio of Ocimum basilicum L.ethyl acetate extract was stronger than that of Ocimum basilicum L.butanol extract(P0.05).CONCLUSION The experiment shows that maximal inhibiting concentration of Ocimum basilicum L.extracts to the growth of RAW264.7 cells reaches 100 μg/mL in relation to intracellular 5-lipoxygenase inhibitor in vitro.
