摘要
目的:探讨维药罗勒不同提取组分在体外对环氧化酶-1(COX-1)的合成物6-keto-PGF1α浓度的影响,间接证明其对COX-1活性的影响,进而找出该药抗血栓的作用机制及其有效部位。方法:将维药罗勒不同提取组分对Ecv304细胞进行不同时间的药物干预,采用MTT法检测活细胞数目,ELISA法测定细胞上清液中6-keto-PGF1α的浓度。结果:不同组分在不同时间内对6-keto-PGF1α的浓度有不同的影响,在短时间内,OBL-正丁醇和OBL-乙醇均能显著增加6-keto-PGF1α的浓度(P<0.01);长时间内,OBL-乙酸乙酯可显著性地降低6-ket-PGF1α的浓度(P<0.05),而OBL-氯仿和OBL-正丁醇中高剂量可显著增加6-keto-PGF1α的浓度(P<0.01)。结论:维药罗勒可能通过抑制TXA2合成酶的活性,激活COX-1的酶活性,最终维持TXA2-PGI2的平衡而产生抗血栓作用,正丁醇提取部位是其发挥激活COX-1酶活性作用的有效部位。
Objective: To study the effects of different polar extract of Uighur Ocimum basilicum L.on COX-1 activity in ECV304 cells in vitro,and find its anti-thrombus mechanism and effective parts.Methods: Ecv304 cells was interfered with different polar extract of Uighur Ocimum basilicum L.in different time.The number of living cells was detected by MTT method.And the concentration of 6-keto-PGF1αin supernatant was analyzed by ELISA method.Results: There are different effects of different polar extract on the concentration of 6-keto-PGF1α in different time.In short,OBL-n-butyl alcohol and OBL-ethanol can significantly increase the concentration of 6-keto-PGF1α(P0.01);a long time,OBL-ethyl acetate can significantly reduce the concentration of 6-keto-PGF1α(P0.05),whereas OBL-chloroform and OBL-n-butyl alcohol in middle and high doses can significantly increase the concentration of 6-keto-PGF1α(P0.01).Conclusion: Uighur Ocimum basilicum L.may inhibit the activity of TXA2 enzyme and activate the activity of COX-1 enzyme.And then the balance between TXA2 and PGI2 will be maintained which can cause anti-thrombus.And the extract part of n-butyl alcohol is one of effective parts when activating the activity of COX-1 enzyme.
