目的探讨GABA 对大鼠黄体细胞孕酮生成的影响及其机制孕酮生成与细胞凋亡的关系方法未成年W i s t ar 雌性大鼠经P M SG hCG 顺序处理后取出双侧卵巢制成黄体细胞悬浮液进行细胞培养放射免疫分析方法测定激素含量3 末端原位标记法检测离体培养黄体细胞自发凋亡结果1 G A BA 抑制黄体细胞基础孕酮的生成及hCG 刺激下的孕酮生成2 F o r s k o l in 促进黄体细胞孕酮的生成GABA 抑制F o r s k o l in 影响下的孕酮生成3 GABA 可促进培养的黄体细胞自发凋亡结论GABA 无论有无hCG
The present experiment was carried out to study the effect of GABA on progesterone secretion and apoptosis by incubated luteal cells of rat in vitro.Methods: I m m a t u r e f e m a l e r a t s w e r e t r e a t e d w i t h P M S G a n d H C G i n d u c e h y p e r o v u l a t i o n f o r a b t a i n i ng l u t e a l c e l l s .T h e p r o d u c t i o n o f p r o g e s t e r o n e w e r e e l e t e m i n e d b y R I A T U N E L t e c h n i c w as u s e d t o d e t e c t t h e a p o t o t i c c e l l s.Results:1.GABA inhibited progesterone production and hCG-induced progesterone production by cultured rat luteal cells.2.GABA inhibited forskolin- induced progesterone production by cultured rat luteal cells.3.GABA enhanced apoptosis of l u t e a l c e l l s.Conclusion:GABA inhibited progesterone production by cultured rat luteal c e l l .T h e e f f e c t o f G A B A m i g h t b e m e d i a t e d b y G A B A r e c e p t o r a n d a d e n y l a t e c y c l a s e -c A MP messenger system.
