摘要
目的研究分化抑制因子-1(ID-1)、血管内皮生长因子(VEGF)、凝血酶敏感蛋白-1(Tsp-1)及微血管密度(MVD)在皮肤基底细胞癌(BCC)及鳞状细胞癌(SCC)中表达的相关性。方法应用免疫组化法检查皮肤BCC及SCC各25例的手术切除标本中ID-1,VEGF,Tsp-1和微血管标记抗体CD34的表达情况,计数肿瘤MVD;应用RT-PCR方法检测ID-1,Tsp-1和VEGF的mRNA表达情况;应用Western blot方法检查ID-1,Tsp-1和VEGF蛋白水平。结果与正常皮肤相比,皮肤SCC和BCC均高表达MVD,ID-1和VEGF,低表达Tsp-1,差异均有统计学意义(P均<0.05)。结论 ID-1和VEGF与皮肤BCC及SCC组织中的血管生成正相关,二者在促进皮肤SCC和BCC血管生成中具有协同作用,并可能在皮肤SCC和BCC的发生及发展过程中起着重要作用。
Objective To explore the express correlation of differentiation inhibiting factor-1 ( ID-1 ), vascular endothe- lial growth factor(VEGF) , thrombin sensitive protein-1 ( Tsp-1 ) and microvascular density (MVD) in basal cell carcinoma (BCC) and cutaneous squamous cell carcinoma (SCC). Methods Immunohistochemistry method was used to detect the expression of ID-1, VEGF, Tsp-1 and CD34 in 25 cases of patients with BCC, and another 25 cases of patients with SCC, then to count the tumor microvessel density (MVD) ; RT- PCR method was used to detect the mRNA expression of ID-1, VEGF, Tsp-1 and CD34; and Western blot method was used to detect the protein level of ID-1, VEGF, Tsp-1 and CD34. Results Compared with nor- mal skin, it shows higher lever expression for MVD,ID-1 and VEGF in the BCC and SCC, while a lower ex- pression for Tsp-1. And the differences were statistically significant (P 〈 0.05 ). Conclusion ID-land VEGF are positively related to the angiogenesis in the BCC and SCC . Both of them create synergies in an- giogenesis. And they could also play an important role in the process of development of BCC and SCC.
