血管内皮生长因子及其受体Flt-1、KDR在胃癌组织中的表达及意义 被引量:7

摘要 目的探讨胃癌组织中血管内皮生长因子(VEGF)及其受体1(Flt-1)、受体2(KDR)表达和微血管密度(MVD)的意义。方法采用免疫组织化学方法检测208例胃癌组织和139例胃正常黏膜组织VEGF、Flt-1、KDR表达和计数MVD。结果胃癌组织中VEGF及其受体Flt-1、KDR的表达明显高于胃正常黏膜组织(P<0.01);胃癌组MVD比胃正常黏膜组度高[(26.19±16.23)/mm2vs(16.02±09.88)个/mm2,P<0.01]。VEGF、Flt-1、KDR表达与肿瘤浸润深度、淋巴转移密切相关(P<0.05)。在VEGF高表达的区域,Flt-1和KDR的表达亦增强,三者呈正相关(P<0.05)。VEGF表达水平与MVD之间也有正相关性(P<0.05)。结论VEGF及其受体Flt-1、KDR在胃癌中广泛表达,与胃癌患者的病理特征和微血管生成关系密切。胃癌组织中VEGF的表达与受体Flt-1、KDR的表达呈正相关;这些结果提示VEGF、Flt-1、KDR在肿瘤组织的血管生成和侵袭转移中可能起重要作用。 Objective To study the MVD and the expression of VEGF,Flt-1 and KDR in Gastric Carcinoma. Methods MVD and the expression of VEGF,Flt-land KDR were examined by immunohistochemical staining in 208 gastric carcinoma specimens and 139 normal tissues. Results The expression of VEGF, Fit-1 and KDR was significantly higher in gastric carcinoma than in normal tissues (P 〈 0.01 ). MVD at intratumoral areas was significantly higher than at normal gastric (26.19±16.23 vs 16.02±09.88/mm2,p 〈 0.01 ). The expression of VEGF,Flt-1 and KDR was correlated with lymph node metastasis and penetrating depth (P 〈 0.05).In the area with high expression of VEGF, there was intensive expression of Fit-1 and KDR. There was significant positive correlation between VEGF, Fit-1, KDR and MVD(P 〈 0.05). Conclusion VEGF, Fit-1 and KDR are widely expressed in GC tissues,and are positively related to pathological features and MVD of GC patients. The expression of VEGF is significantly correlated with Fit-1, KDR. These results suggest that VEGF, Flt- 1 and KDR have an important role in angiogengesis and metastasis of gastric carcinoma.
