摘要
目的:探讨雌激素受体基因(ER)态性与宁夏地区回族、汉族人群慢性牙周炎(CP)易感性关系,并分析ER基因的基因型和等位基因频率在回、汉人群中的分布是否有差异。方法:收集慢性牙周炎患者和健康对照者静脉血,用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对受检者进行ER-α/XbaⅠ和ER-α/PvuⅡ两个多态性位点的基因型检测,计算基因型频率和等位基因频率,并进行病例对照分析。结果:汉族、回族CP组和对照组在ER-α/XbaⅠ基因型和等位基因频率分布上均无差异;汉族CP组和对照组在ER-α/PvuⅡ基因型和等位基因频率分布上均无差异,回族CP组和对照组在ER-α/PvuⅡ基因型的分布上有显著性差异(P<0.05),在基因频率分布上无差异;汉族CP组和回族CP组在ER-α/XbaⅠ基因型和等位基因频率分布上均无差异,在ER-α/PvuⅡ基因型和等位基因频率的分布上均有显著性差异(P<0.05)。结论:ER-α/XbaⅠ位点多态性与宁夏地区回族、汉族人群慢性牙周炎的易感性无相关关系。ER-α/PvuⅡ位点基因多态性可能与宁夏地区回族以及汉族人群慢性牙周炎的易感性有关,其基因型的分布可能为回族慢性牙周炎的易感因子。
Objective： To determine the association between estrogen receptors gene polymorphisms and chronic periodontitis in Hui and Han populations from Ningxia,and to analyze the distributions of ER genotypes and alleles frequency between the two nationalities.Methods： We collected peripheral venous blood from participants,including 135 patients with chronic periodontitis（CP） and 139 healthy controls in Han nationality;120 patients with chronic periodontitis and 126 healthy controls in Hui nationality.The genotypes of ER-α/XbaⅠand ER-α/PvuⅡ were investigated by PCR-RFLP respectively.We evaluated the genotypes and alleles frequency of ER-α/XbaⅠand ER-α/PvuⅡ from all the subjects and analyzed that whether there are associations between ER gene polymorphisms and CP by case-control methods.Results： There was no statistically significant difference in the frequencies of ER-α/XbaⅠ genotypes and alleles between the CP patients and healthy controls in Hui and Han nationality.There was no statistically significant difference in the frequencies of ER-α/PvuⅡ genotypes and alleles between the CP patients and healthy controls in Han nationality.There was statistically significant difference in the frequencies of ER-α/PvuⅡ genotypes（P0.05）,no significant difference alleles between the CP patients and healthy controls in Hui nationality.The difference in the frequencies of ER-α/XbaⅠ genotypes and alleles was no statistic significance between the Hui CP group and Han CP group.The difference in the frequencies of ER-α/PvuⅡ genotypes and alleles has statistic significance between the Hui CP group and Han CP group（P0.05）.Conclusion： The polymorphism of the ER-α/XbaⅠ was not associated with the chronic periodontitis in Hui and Han nationality.The polymorphism of the ER-α/PvuⅡ was associated with the chronic periodontitis in Hui and Han nationality,the ER-α/PvuⅡ genotype may be a susceptible indicator for chronic periodontitis in Hui nationality.
