摘要
【目的】探讨葶苈大枣泻肺汤对肺癌小鼠恶性胸腔积液及水通道蛋白1(AQP1)的影响。【方法】选用C57BL/6J小鼠建立肺癌动物模型,分为肿瘤组和中药组,并设立正常对照组,记录各组胸水量,并采用免疫组织化学及Real-time PCR法测定AQP1及AQP1 mRNA水平。【结果】肿瘤组小鼠胸腔内见血性积液及肿瘤结节生长,中药组小鼠胸水量较肿瘤组显著减少,壁层胸膜AQP1水平及mRNA表达均较肿瘤组显著降低(P<0.05或P<0.01)。【结论】葶苈大枣泻肺汤可以抑制肺癌小鼠壁层胸膜AQP1及mRNA表达,减少恶性胸水。
Objective To reveal the effect of Tingli Dazao Xiefei Decoction(TDXD) on aquaporin 1(AQP1) and malignant pleural effusion of mice with lung cancer.Methods We established the C57BL/6J mice model of lung cancer,and the model mice were divided into the tumor model group and TDXD group.And a normal control group was also set up.The amount of malignant pleural effusion was recorded,and the levels of AQP1 and its mRNA expression in parietial pleura were detected by using immunohistochemistry and real-time PCR.Results Bloody effusion and tumor nodules appeared in the pleural cavity of tumor model group.In TDXD group,the amount of pleural effusion was obviously decreased,and AQP1 level and its mRNA expression in parietial pleura were markedly decreased(P〈0.05 or P〈0.01).Conclusion TDXD shows an effect on decreasing AQP1 level and its mRNA expression in parietial pleura,and on reducing the amount of malignant pleural effusion in mice with lung cancer.
