摘要
目的:观察肾乐方及其拆方对阿霉素肾病大鼠P-选择素的干预作用,进一步探讨肾乐方组方中不同治法的作用机制及其配伍意义。方法:实验动物随机分为6组:正常对照组、模型对照组、肾乐方组、益气养阴组、清热利湿组、活血化瘀组。建立阿霉素肾病模型(尾静脉注射,剂量6 mg/kg),测定大鼠尿蛋白定量、血清白蛋白(ALB)、胆固醇(ChoL)、肌酐(Scr)含量,以酶联免疫吸附法、S-P免疫组化法分别检测血清、肾组织P-选择素的表达,并进行电镜下肾脏组织形态学观察。结果:与正常组比较,阿霉素肾病模型大鼠尿蛋白定量明显升高,血清AlB水平明显降低,ChoL、Scr水平明显升高,血清、肾组织P-选择素表达水平升高(P<0.05,P<0.01),电镜肾脏超微结构观察显示肾小球足突重度融合。肾乐方及其拆方可减少阿霉素肾病大鼠尿蛋白的丢失,改善血液生化指标,降低血清、肾组织P-选择素的表达(P<0.05,P<0.01),以全方组效果最为显著。结论:肾乐方及其拆方可能通过降低阿霉素肾病大鼠增高的P-选择素表达,对肾脏起到保护作用。各拆方组之间具有显著的协同增效作用。
Objective：To investigate the effects of Shenle Decoction and its components on P-selectin in adriamycin nephrosis rats.Methods：The adriamycin nephrosis model rats were induced by ADR venoclysis through caudal vein.The rats were randomly divided into six groups： the normal control group,the model control group,Shenle Decoction group,the Yiqiyangyin Chinese herbs group,the Qingrelishi Chinese herbs group,the Huoxuehuayu Chinese herbs group.Different treatments were given to groups for 4 weeks.We observed the amount of protein in urine,determined the levels of ALB,ChoL,Scr and P-selectin in serum and the levels of P-selectin in kidney tissue,and observed morphologic change in kidneys.Results：Compared with the normal group,the levels of P-selectin in serum and kidney tissue of rats in the model group were increased（P〈0.01）.Shenle Decoction and its components could decrease the amount of protein in urine and the levels of ChoL,Scr and P-selectin in serum and the levels of P-selectin in kidney tissue（P〈0.05,P〈0.01）,increase the levels of ALB in serum.Conclusion：Shenle Decoction and its components have obvious protective effect on adriamycin nephrosis rats,can decrease the levels of P-selectin in serum and kidney tissue,and prevent the further development of adriamycin nephrosis.All these prescriptions have the cooperative effects.It provides theoretic proofs in treating and preventing nephrotic syndrome.
