摘要
目的观察益气活血中药配伍双联抗血小板治疗对急性心肌梗死(AMI)后大鼠血小板功能和血栓形成的影响。方法 59只SD大鼠随机选12只设为假手术组,其余大鼠结扎前降支冠状动脉建立实验性AMI模型,36只造模成功大鼠随机分为模型组、双抗组和益气活血+双抗组各12只。双抗组给予阿司匹林肠溶片9mg/(kg·d)和硫酸氢氯吡格雷片6.75mg/(kg·d)灌胃,益气活血+双抗组在双抗组的基础上加给予心悦胶囊162mg/(kg·d)+复方川芎胶囊400mg/(kg·d)灌胃,模型组和假手术组给予等量蒸馏水,连续灌胃28天。测定大鼠体内血栓形成时间和体外血栓形成长度、湿重、干重,检测凝血指标、血小板最大聚集率(PAgT)、血栓素B2(TXB2)和6酮-前列腺素F1α(6-keto-PGF1α)含量,计算心室腔面积与心脏横切面积比值。结果与假手术组比较,模型组大鼠颈动脉体内血栓形成时间缩短,体外血栓形成长度、湿重和干重增加,活化部分凝血酶时间(APTT)缩短,纤维蛋白原(FIB)含量增加,6-keto-PGF1α含量降低,心室腔面积和心脏横切面积比值增加(P<0.05或P<0.01);与模型组比较,益气活血+双抗组大鼠体内血栓形成时间显著延长,体外血栓长度、湿重、干重显著降低,凝血指标改善,PAgT下降,6-keto-PGF1α含量升高,心室腔面积和心脏横切面积比值降低(P<0.05或P<0.01)。结论益气活血中药配伍双联抗血小板治疗可进一步抑制AMI大鼠颈动脉动脉血栓形成,且不增加出血风险。
Objective To observe the effect of qi-supplementing blood-quickening medicinal combined with dual antiplatelet therapy on the platelet function and thrombosis in acute myocardial infarction (AMI) rats. Methods Twelve rats among 59 SD rats were randomly selected as the sham operation group and other rats were established AMI models by ligation of the left anterior descending coronary artery. Thirty six successfully modeling rats were randomized into the model group, dual antiplatelet therapy group and qi-supplementing blood-quickening medicinal combined with dual antiplatelet therapy group, with 12 in each. The dual antiplatelet therapy group was gavaged with 9 mg/(kg·d) aspirin and 6.75 mg/(kg·d) clopidogrel. The qi-supplementing blood-quickening medicinal combined with dual antiplatelet therapy group was gavaged with 162 mg/ (kg·d) Xinyue Capsule and 400 mg/(kg·d) Compound Chuanxiong Capsule on the basis of the dual antiplatelet therapy group. The model group and sham operation group were gavaged with distilled water for 28 days. The thrombus formation time, length, wet weight and dry weight of thrombosis in vitro, coagulation, platelet aggregation rate (PAgT), thromboxane B2 (TXB2) and 6-keto-prostaglandin F1a (6-keto-PGF1a) was detected. The cardiac ventricular area and cross-sectional area ratio was calculated. Results Comparing with the sham operation group, the thromhus formation time of carotid artery in vivo and activated partial thromboplastin time (APTT) were significantly shortened, the length, wet weight and dry weight of thrombosis in vitro, fibrinogen (FIB) content and cardiac ventricular area and cross-sectional area ratio were significantly increased and 6-keto-PGF1a was significantly decreased in the model group (P〈0.05 or P〈0.01). Comparing with the model group, the thrombus formation time in vivo was significantly prolonged, the length, wet weight and dry weight of thrombosis in vitro, PAgT and cardiac ventricular area and cross sectional area
