摘要
目的研究温阳振衰方对阿霉素所致大鼠慢性心力衰竭模型心功能、心脏结构及心脏基质金属蛋白酶2和9的影响,探讨温阳振衰方防治慢性心力衰竭的作用机理。方法采用冷冻配合腹腔注射阿霉素的方法制备慢性心衰模型,随机分为心力衰竭模型组、阳性对照组(卡托普利组)、温阳振衰方组,另有空白组10只。各组相应处理4周后,检测并比较各组心脏彩超结果、心脏组织基质金属蛋白酶(MMP)2和9的变化。结果温阳振衰方组与卡托普利组的大鼠与心衰模型组比较心功能(彩超结果)均得到改善,LVEF、FS水平均较心衰模型组明显升高,LVEDD、LVESD水平均较心衰模型组明显降低;MMP-2和MMP-9明显下降;温阳振衰方组与卡托普利组比较心功能指标FS、LVESD无显著差异;其他各指标改善,温阳振衰方组优于卡托普利组。结论温阳振衰方及卡托普利均能改善心功能,能够降低基质金属蛋白酶2和9的水平,从而改变心脏细胞外基质的环境,进而延缓心肌纤维化进展。温阳振衰方降低基质金属蛋白酶2和9及改善心功能的作用稍优于卡托普利。
Objective TO study the effects of Wenyang Zhenshuai decoction(WZD) on heart function,heart structure, MMP-2 and MMP-9 with chronic congestive heart failure. , and to explore the rapeutic mechanism of Wenyang Zhenshuai decoction (WZD)with chronic congestive heart failure. Methods Select 10 rabbits randomly for normal group,and the left 40 mouse which will be model with chronic congestive heart failure by freezing 2 hours everyday and injecting Azithromycin once a week for 5 weeks,then the rabbits will be divided into three groups: model group, control group (Captopril group), Wenyang Zhenshuai group. After 4 weeks, UCG, cardiac tissue inflammation factor matrix metalloproteinase (MMP) 2 and 9 were tested and compared. Results After treatment: Compared with model group, the heart function of Captopril group and Wenyang Zhenshuai group were improved significantly,the levels of MMP-2 and MMP-9 were decreased;Compared with the Captopril group, There were not significant differences in heart function (FS,LVEDD) of Wenyang Zhenshuai group. There was difference in other index between the Wenyang Zhenshuai group and Captopril group . Conclusion Wenyang Zhenshuai group and Captopril group can improve the function of the heart,regulate the level of the MMP(2,9) which changes the Cardiac extracellular matrix environment, and delay the myocardial fibrosis progression. And WZD is a little better than Captopril.
